| Application Notes / Precautions | For whole-cell lysis, use 0.5–1.0% NP-40 in lysis buffer for 30 minutes on ice with gentle agitation. For nuclear extract preparation, use 0.1% NP-40 for cytoplasmic membrane lysis followed by nuclear pellet extraction. NP-40 interferes with SDS-PAGE at concentrations >1%; dilute lysates accordingly before loading. Always include fresh protease and phosphatase inhibitors in lysis buffer. |
