Product Name Guanidinium Isothiocyanate Lysis Solution, 4 M
Catalog No. ATR-SDS-0013
Description A concentrated chaotropic lysis reagent containing 4 M guanidinium isothiocyanate for the rapid denaturation of proteins and inactivation of RNases in RNA extraction from cells and tissues.
Intended Use Primary lysis and homogenization reagent in single-step RNA isolation methods, immediately denaturing cellular proteins and inactivating endogenous and environmental RNases upon contact.
Principle / Technology Guanidinium isothiocyanate is a powerful chaotropic agent that disrupts hydrogen bonding in proteins, denaturing all cellular proteins including RNases within seconds of contact; beta-mercaptoethanol reduces disulfide bonds to ensure complete denaturation.
Detection Method RNA integrity by Bioanalyzer RIN score, A260/A280 ratio, RT-PCR functional validation
Sample Type Cultured cells, fresh and frozen tissues, bacterial pellets, yeast
Performance Range / Specifications RNA yield 5–50 µg per 10^6 cells; RIN score >8.0 for properly handled fresh samples
Sensitivity / LOD N/A
Specificity N/A
Reaction Conditions / Protocol Add 1 mL per 10^7 cells or 100 mg tissue; homogenize immediately; proceed to organic extraction or silica column purification.
Components / Formulation Guanidinium isothiocyanate 4 M, sodium citrate pH 7.0 25 mM, sodium N-lauroylsarcosine 0.5%, beta-mercaptoethanol 100 mM (or DTT 10 mM)
Storage Conditions 15–30°C, protect from light; add reducing agent fresh before use
Shelf Life 12 months (without reducing agent)
Package Specifications 100 mL, 500 mL
Product Form Liquid
Quality Control RNase inactivation validation, RNA integrity testing with HeLa cell standard, pH verification
Key Features Instant RNase inactivation; standard for RNA extraction; compatible with organic and column purification; validated for tissues and cells
