| Product Name | WS Bst DNA Polymerase (40 U/μL, Warm-Start) |
| Catalog No. | IVDRM-0271 |
| Description | WS Bst DNA Polymerase (40 U/μL, Warm-Start) is a warm-start isothermal amplification polymerase derived from Bacillus stearothermophilus DNA Polymerase I. The enzyme possesses 5'→3' DNA polymerase activity and strong strand-displacement activity but lacks 5'→3' exonuclease activity, making it suitable for DNA strand-displacement reactions, loop-mediated isothermal amplification (LAMP), and rapid sequencing. A reversible aptamer-modification technology completely blocks enzyme activity at room temperature, preventing non-specific amplification and allowing reactions to be set up at ambient temperature—ideal for high-throughput and large-scale screening workflows. No separate activation step is required: full enzyme activity is released within 1 minute at temperatures above 60°C. |
| Intended Use | Warm-start isothermal amplification (LAMP/RT-LAMP) and DNA strand-displacement reactions; suitable for room-temperature reaction setup in high-throughput and large-scale screening applications. |
| Principle / Technology | Aptamer-modified warm-start Bst DNA polymerase: a reversible aptamer blocks polymerase activity at room temperature, suppressing non-specific amplification during reaction setup; upon heating above 60°C, the aptamer dissociates and full enzyme activity is restored within 1 minute. The enzyme catalyzes 5'→3' DNA synthesis with strong strand-displacement activity (5'→3' exonuclease deficient) for isothermal amplification. |
| Detection Method | Isothermal amplification (LAMP, RT-LAMP); compatible with fluorescent dye-based detection formats |
| Sample Type | DNA templates (RT-LAMP when combined with reverse transcriptase) |
| Performance Range / Specifications | Concentration: 40 U/μL Activity completely blocked at room temperature, preventing non-specific amplification Full enzyme activity released within 1 minute at >60°C; no separate activation step required Delivers lower background signal and delayed non-specific amplification onset compared with unmodified Bst polymerase in room-temperature setups |
| Specificity | Aptamer-mediated warm-start inhibition at room temperature reduces background signal and suppresses non-specific amplification, improving assay specificity. |
| Reaction Conditions / Protocol | Reactions can be assembled at room temperature. Amplification is performed at standard LAMP temperatures (60–68°C); enzyme activity is fully released within 1 minute above 60°C, with no separate activation step. |
