Description Alta DiagnoTech alpha-glucosidase is a pale-yellowish white amorphous enzyme powder for amylase biochemical reagent development.
Intended Use Enzyme raw material for amylase biochemical assay reagents.
Principle / Technology The enzyme releases p-nitrophenol from a chromogenic alpha-D-glucoside substrate; activity is calculated from the absorbance-change rate at 405 nm.
Detection Method Kinetic p-nitrophenol absorbance assay at 405 nm
Performance Range / Specifications Specific activity >=10 U/mg; assay linear range 100-800 U/L; recommended sample range 200-400 U/L.
Reaction Conditions / Protocol Prepare the assay reagent by dissolving 13.6 g potassium dihydrogen phosphate in 800 mL water, adjusting to pH 7.0 at 25 degrees C with KOH, adding 1.50 g 4-nitrophenyl-alpha-D-glucopyranoside and 1 g sodium azide, adjusting to pH 7.00 +/- 0.05 and bringing to 1 L. Preincubate 0.900 mL assay reagent for 60 seconds at 37 degrees C, add 0.018 mL enzyme sample, mix, and after 1 minute determine the absorbance-change rate from 180 to 240 seconds at 405 nm. Activity in U/L equals Delta A/min x 2,818 x dilution factor.
