Description A glycerol-free warm-start Bst DNA polymerase engineered for high-sensitivity isothermal amplification and freeze-dried reagent development.
Intended Use LAMP, RT-LAMP and related isothermal amplification assays.
Principle / Technology The enzyme retains 5′→3′ polymerase activity and strong strand-displacement activity while lacking 5′→3′ exonuclease activity. An auxiliary factor increases LAMP sensitivity, and warm-start control suppresses activity below 50°C.
Detection Method Endpoint, colorimetric or real-time fluorescent isothermal amplification.
Sample Type Purified nucleic acids and compatible crude lysates, including saliva and nasal or throat swab lysates.
Performance Range / Specifications 1,600 U or 8,000 U; reported sensitivity improvement of approximately 10- to 100-fold in LAMP applications; supports room-temperature setup.
Sensitivity / LOD Designed for improved detection of low-copy templates.
Specificity Warm-start inhibition below 50°C reduces nonspecific amplification during reaction setup.
Reaction Conditions / Protocol Combine with isothermal amplification buffer, magnesium, dNTPs, assay primers and template. Incubate above 50°C using conditions optimized for the target assay.
