| Product Name | Uracil DNA Glycosylase (UDG/UNG), Heat-Labile (1 U/μL) |
| Catalog No. | IVDRM-0248 |
| Description | Heat-labile Uracil DNA Glycosylase (UDG/UNG) catalyzes the hydrolysis of the N-glycosidic bond between the uracil base and the sugar-phosphate backbone of uracil-containing DNA, releasing free uracil. Compared with conventional UDG, this heat-labile enzyme avoids the degradation of dU-containing amplification products at room temperature caused by residual activity that may persist after inactivation of ordinary UDG. The enzyme works at room temperature, is temperature-sensitive and is easily inactivated. |
| Intended Use | Prevention of carryover (aerosol) contamination from dU-containing PCR amplification products in dUTP-based PCR/qPCR systems Removal of uracil bases from single-stranded or double-stranded DNA |
| Principle / Technology | UDG excises uracil from DNA by hydrolyzing the N-glycosidic bond linking the uracil base to the deoxyribose-phosphate backbone, generating apyrimidinic (AP) sites. When PCR is performed with dUTP instead of dTTP, contaminating carryover amplicons from previous reactions are degraded by a UDG pre-treatment step, while the natural thymine-containing template remains unaffected. |
| Performance Range / Specifications | Low nuclease background: free of exonuclease, nicking endonuclease and RNase contamination Broad application: removes uracil bases from single-stranded or double-stranded DNA Heat-labile: inactivated at 95 °C for 5-10 minutes Compatible with RT-qPCR systems: no impact on the detection system even at high input (2 U per 20 uL reaction) |
| Specificity | Specifically excises uracil bases from single-stranded and double-stranded DNA; does not act on natural thymine-containing DNA templates. |
| Reaction Conditions / Protocol | Active at room temperature. A UDG pre-treatment step is performed before amplification in dUTP-based PCR/qPCR systems; the enzyme is then inactivated during the initial high-temperature step. |
| Storage Conditions | Store at -25 °C to -15 °C. |
| Shelf Life | 2 years when stored at -25 °C to -15 °C. |
| Package Specifications | 100 U 500 U 10,000 U 100,000 U 500,000 U |
| Product Form | Liquid |
| Quality Control | Free of exonuclease, nicking endonuclease and RNase contamination. |
| Concentration | 1 U/uL |
| Expiration Date / Stability | Stable for 2 years at -25 °C to -15 °C. |
| Compatibility | Compatible with RT-qPCR reaction systems; no impact on the detection system at high input (2 U per 20 uL reaction). |
| Application Notes / Precautions | Unlike conventional UDG, this heat-labile enzyme eliminates the risk of residual activity degrading dU-containing amplification products at room temperature after the reaction. Use in dUTP-based amplification systems for carryover contamination control. |
| Recognition Site | Uracil bases in single-stranded or double-stranded DNA (cleaves the N-glycosidic bond between uracil and the sugar-phosphate backbone) |
| Heat Inactivation | 95 °C for 5-10 minutes |
