| Product Name | Uracil DNA Glycosylase (UDG), Heat-Labile (10 U/μL, Glycerol-Free) |
| Catalog No. | IVDRM-0260 |
| Description | Heat-Labile Uracil DNA Glycosylase (UDG) catalyzes the hydrolysis of the N-glycosidic bond between the uracil base and the sugar-phosphate backbone of uracil-containing DNA strands, releasing free uracil. Compared with conventional UDG, this heat-labile enzyme avoids the residual activity that may persist after inactivation of regular UDG and degrade dU-containing amplification products at room temperature. The enzyme is active at room temperature, is highly temperature-sensitive, and is easily inactivated. With extremely low glycerol content it can be lyophilized, and its high activity (10 U/μL) facilitates reaction system optimization. It is widely used to prevent aerosol carryover contamination of PCR products. |
| Intended Use | Prevention of carryover (aerosol) contamination in PCR, qPCR, and isothermal amplification workflows by degrading uracil-containing DNA from previous reactions; raw material enzyme for molecular diagnostic reagent formulation, including lyophilized systems. |
| Principle / Technology | UDG catalyzes hydrolysis of the N-glycosidic bond linking uracil bases to the deoxyribose-phosphate backbone in uracil-containing DNA, releasing free uracil and generating abasic sites that prevent the DNA from serving as an amplification template. The heat-labile design allows the enzyme to act at room temperature and to be readily inactivated by heating, eliminating residual activity that could degrade newly generated dU-containing amplicons. |
| Sample Type | Uracil-containing (dU-containing) DNA |
| Performance Range / Specifications | Extremely low glycerol content; suitable for lyophilization. High activity (10 U/μL), convenient for reaction system optimization. Active at room temperature and easily heat-inactivated. |
| Specificity | Specifically hydrolyzes the N-glycosidic bond of uracil in DNA strands. |
| Reaction Conditions / Protocol | The enzyme is active at room temperature and can be applied during reaction setup or a pre-incubation step to eliminate carryover contaminants before amplification. |
| Components / Formulation | Glycerol-free (extremely low glycerol) heat-labile UDG enzyme solution at 10 U/μL. |
| Storage Conditions | Store at 2-8°C. |
| Shelf Life | 1 year when stored at 2-8°C. |
| Package Specifications | 100 U 500 U 10000 U 30000 U |
| Product Form | Glycerol-free liquid formulation, 10 U/μL; suitable for lyophilization. |
| Concentration | 10 U/μL |
| Expiration Date / Stability | Valid for 1 year when stored at 2-8°C. |
| Compatibility | Compatible with PCR/qPCR systems using dUTP and with lyophilized reagent formulations. |
| Application Notes / Precautions | Use to prevent aerosol carryover contamination of amplification products. Because the enzyme is heat-labile, ensure an adequate heat inactivation step so that no residual activity degrades dU-containing amplification products at room temperature. The glycerol-free formulation allows direct use in lyophilized reagent preparation. |
| Recognition Site | Uracil bases in uracil-containing (dU-containing) DNA strands |
| Heat Inactivation | Heat-labile: highly temperature-sensitive and readily inactivated by heat treatment, eliminating residual activity toward dU-containing amplification products (specific inactivation temperature/time not specified in source material). |
