| Product Name | Super Bst DNA Polymerase (120 U/μL) |
| Catalog No. | IVDRM-0270 |
| Description | Super Bst DNA Polymerase (120 U/μL) is an engineered derivative of Thermophilic Geobacillus sp. DNA Polymerase I in which the 5'→3' exonuclease activity has been removed. The enzyme exhibits strong 5'→3' DNA polymerase activity, robust strand-displacement activity, and tolerance to dUTP, making it well suited for contamination-controlled isothermal amplification reactions such as LAMP and CPA. Its enhanced thermostability supports amplification at up to 70°C, improves performance with low-concentration templates, shortens time-to-result, and provides a foundation for developing hot-start Bst formulations that further increase detection specificity. |
| Intended Use | Suitable for a variety of isothermal amplification reactions, including LAMP, CPA, and RCA; supports 70°C LAMP and, in combination with a thermostable MMLV reverse transcriptase, 70°C RT-LAMP. |
| Principle / Technology | Engineered Bst-family DNA polymerase with strong 5'→3' polymerase and strand-displacement activity (5'→3' exonuclease deficient), enabling rapid isothermal amplification of DNA targets at a constant temperature, including elevated-temperature (70°C) LAMP formats. dUTP tolerance enables UDG-mediated carryover contamination control. |
| Detection Method | Isothermal amplification (LAMP, CPA, RCA, RT-LAMP) |
| Sample Type | DNA templates, including low-concentration samples |
| Performance Range / Specifications | Concentration: 120 U/μL Compatible with LAMP, CPA, RCA and other isothermal amplification reactions High thermostability: enables LAMP at 70°C, or 70°C RT-LAMP when paired with a thermostable MMLV reverse transcriptase |
| Sensitivity / LOD | Supports amplification of low-concentration templates; increasing reaction temperature to 70°C significantly shortens Ct values and further improves assay sensitivity. |
| Specificity | Elevated-temperature amplification (70°C) increases detection specificity; dUTP tolerance allows use in UDG-based anti-contamination systems. |
| Reaction Conditions / Protocol | Typical LAMP reaction temperature: 65–70°C. For RT-LAMP at 70°C, combine with a thermostable MMLV reverse transcriptase. |
