Product Name Robust Hot-Start Taq DNA Polymerase (20 U/μL, Inhibitor-Tolerant)
Catalog No. IVDRM-0266
Description This hot-start DNA polymerase is dual-blocked with two proprietary antibodies that suppress both the 5'→3' polymerase activity and the 5'→3' exonuclease activity of Taq DNA polymerase. A 30-second heating step at the pre-denaturation temperature fully inactivates the antibodies, releasing both polymerase and exonuclease activities. The dual-blocking design prevents non-specific amplification caused by mispriming or primer dimers and also inhibits fluorescence signal decay caused by probe degradation, improving the stability of in vitro diagnostic reagents during shipping and room-temperature handling. The enzyme shows strong tolerance to inhibitors from blood, swabs, sputum, and feces, as well as phenolic and alcoholic residues from sample pretreatment, supporting direct amplification from crude samples.
Intended Use For the development of qPCR/PCR-based in vitro diagnostic assays, including direct amplification from crude or inhibitor-rich samples such as blood, swab eluates, sputum, and feces.
Principle / Technology Dual-antibody hot-start technology: two blocking antibodies suppress the 5'→3' polymerase and 5'→3' exonuclease activities of Taq DNA polymerase at low temperatures; both activities are restored after a 30-second incubation at the pre-denaturation temperature.
Detection Method Real-time PCR (qPCR) / probe-based PCR
Sample Type Crude or minimally processed samples: swab eluates, plasma/whole blood, sputum, feces; templates containing phenol or alcohol residues from sample pretreatment
Performance Range / Specifications Antibody-modified hot start: activation at 95°C for 30 sec-1 min
High sensitivity and strong specificity
Tolerates inhibitors from blood, swab, sputum, and feces, as well as phenolic and alcoholic residues from sample pretreatment
Supports direct amplification of swab samples processed in direct-lysis buffer, outperforming comparable market products
Tolerates plasma at up to approximately 30% final reaction concentration in direct qPCR amplification
Sensitivity / LOD High sensitivity for qPCR detection; supports direct amplification from crude swab lysates
Specificity Dual antibody blocking effectively prevents non-specific amplification from mispriming and primer dimers, and suppresses probe degradation-related fluorescence signal loss
Reaction Conditions / Protocol Hot-start activation at 95°C for 30 sec-1 min, followed by standard PCR/qPCR cycling.
