| Product Name | RNase A (100 mg/mL) |
| Catalog No. | IVDRM-0184 |
| Description | Ribonuclease A (RNase A) is a single-chain polypeptide containing four disulfide bonds, with a molecular weight of approximately 13.7 kDa. As an endoribonuclease, it specifically degrades RNA at cytosine (C) and uracil (U) residues. RNase A shows the highest activity on single-stranded RNA and remains active under a wide range of reaction conditions: at low salt concentrations (0-100 mM NaCl) it cleaves single-stranded RNA, double-stranded RNA, and the RNA strand of RNA-DNA hybrids, whereas at high salt concentrations (≥0.3 M) it cleaves single-stranded RNA only. The most common application of RNase A is the removal of RNA during plasmid DNA or genomic DNA preparation; contaminating DNase activity is a key concern in such preparations and is traditionally inactivated by boiling in a water bath. This product is also suitable for RNase protection assays, RNA sequence analysis, and other molecular biology applications. The product is supplied as a solution at 100 mg/mL; the recommended working concentration is 1-100 μg/mL, depending on the application. |
| Intended Use | For removal of RNA during plasmid DNA or genomic DNA preparation; also suitable for RNase protection assays, RNA sequence analysis, and other molecular biology experiments. |
| Principle / Technology | RNase A is an endoribonuclease that specifically degrades RNA at cytosine (C) and uracil (U) residues. It cleaves the phosphodiester bond formed between the 5'-ribose of one nucleotide and the phosphate group attached to the 3'-ribose of an adjacent pyrimidine nucleotide, hydrolyzing the 2',3'-cyclic phosphate intermediate to the corresponding 3'-nucleoside phosphate (for example, pG-pG-pC-pA-pG is cleaved by RNase A to yield pG-pG-pCp and A-pG). |
| Sample Type | Single-stranded RNA; under low-salt conditions also double-stranded RNA and the RNA strand of RNA-DNA hybrids. |
| Performance Range / Specifications | Highest activity on single-stranded RNA; active under a wide range of reaction conditions. Unit activity is defined in Kunitz units at 25°C and pH 5.0. |
| Specificity | Specifically degrades RNA at cytosine (C) and uracil (U) residues of single-stranded RNA; at high salt concentrations (≥0.3 M NaCl), cleaves single-stranded RNA only. |
| Reaction Conditions / Protocol | Active under a wide range of conditions. At low salt concentrations (0-100 mM NaCl), cleaves single-stranded RNA, double-stranded RNA, and the RNA strand of RNA-DNA hybrids; at high salt concentrations (≥0.3 M), cleaves single-stranded RNA only. Recommended working concentration: 1-100 μg/mL, depending on the application. |
