Product Name RNase A (10 mg/mL)
Catalog No. IVDRM-0183
Description Ribonuclease A (RNase A) is a single-chain polypeptide containing four disulfide bonds, with a molecular weight of approximately 13.7 kDa. As an endoribonuclease, it specifically degrades RNA at cytosine (C) and uracil (U) residues. RNase A shows the highest activity on single-stranded RNA and remains active under a wide range of reaction conditions: at low salt concentrations (0-100 mM NaCl) it cleaves single-stranded RNA, double-stranded RNA, and the RNA strand of RNA-DNA hybrids, whereas at high salt concentrations (≥0.3 M) it cleaves single-stranded RNA only. The most common application of RNase A is the removal of RNA during plasmid DNA or genomic DNA preparation. Because contaminating DNase activity is a key concern in such preparations, DNase is traditionally inactivated by boiling in a water bath. This product is free of DNase and protease, so no heat treatment is required before use. It is also suitable for RNase protection assays, RNA sequence analysis, and other molecular biology applications. The product is supplied as a solution at 10 mg/mL; the recommended working concentration is 1-100 μg/mL, depending on the application.
Intended Use For removal of RNA during plasmid DNA or genomic DNA preparation; also suitable for RNase protection assays, RNA sequence analysis, and other molecular biology experiments.
Principle / Technology RNase A is an endoribonuclease that specifically degrades RNA at cytosine (C) and uracil (U) residues. It cleaves the phosphodiester bond formed between the 5'-ribose of one nucleotide and the phosphate group attached to the 3'-ribose of an adjacent pyrimidine nucleotide, hydrolyzing the 2',3'-cyclic phosphate intermediate to the corresponding 3'-nucleoside phosphate (for example, pG-pG-pC-pA-pG is cleaved by RNase A to yield pG-pG-pCp and A-pG).
Sample Type Single-stranded RNA; under low-salt conditions also double-stranded RNA and the RNA strand of RNA-DNA hybrids.
Performance Range / Specifications Highest activity on single-stranded RNA; active under a wide range of reaction conditions. Unit activity is defined in Kunitz units at 25°C and pH 5.0.
Specificity Specifically degrades RNA at cytosine (C) and uracil (U) residues of single-stranded RNA; at high salt concentrations (≥0.3 M NaCl), cleaves single-stranded RNA only.
Reaction Conditions / Protocol Active under a wide range of conditions. At low salt concentrations (0-100 mM NaCl), cleaves single-stranded RNA, double-stranded RNA, and the RNA strand of RNA-DNA hybrids; at high salt concentrations (≥0.3 M), cleaves single-stranded RNA only. Recommended working concentration: 1-100 μg/mL, depending on the application.
Components / Formulation Supplied in 50 mM Tris-HCl (pH 7.4) with 50% (v/v) glycerol.
Storage Conditions Store at -25°C to -15°C. Storage buffer: 50 mM Tris-HCl (pH 7.4) with 50% (v/v) glycerol.
Shelf Life 2 years.
Package Specifications 1 mL
Product Form Liquid (solution).
Quality Control Free of DNase and protease; no heat treatment is required before use.
Concentration 10 mg/mL
Activity / Unit Definition One unit is defined as the amount of enzyme that hydrolyzes RNA at a rate constant k equal to unity (Kunitz units) at 25°C and pH 5.0.
Molecular Weight Approximately 13.7 kDa
Source / Origin Bovine pancreas.
Shipping Conditions Shipped with ice packs.
Application Notes / Precautions Sourced from bovine pancreas.
Boiling used to inactivate contaminating DNase does not affect RNase activity, because RNase A is stable at high temperatures for short periods.

For research use only, not for clinical use.
