| Product Name | Reverse Transcriptase V (Thermostable) |
| Catalog No. | IVDRM-0242 |
| Description | This product is a new-generation reverse transcriptase derived from an RNase H-deficient M-MLV (H-) reverse transcriptase through genetic engineering. Compared with its M-MLV (H-) predecessor, its thermostability is greatly improved, tolerating reaction temperatures of up to 60 °C, making it suitable for reverse transcription of RNA templates with complex secondary structures. The enzyme also exhibits enhanced template affinity, making it well suited for reverse transcription of limited templates and low-copy genes. Its ability to synthesize full-length cDNA is improved, enabling amplification of cDNA up to 10 kb. |
| Intended Use | Raw material enzyme for RT-PCR and RT-qPCR reagent development, including assays targeting structured RNA templates, low-abundance templates, and low-copy genes. |
| Principle / Technology | Genetically engineered RNase H-deficient M-MLV reverse transcriptase with enhanced thermostability and template affinity for efficient cDNA synthesis at elevated temperatures. |
| Detection Method | RT-PCR / RT-qPCR |
| Sample Type | RNA templates, including RNA with complex secondary structures and low-copy or low-abundance RNA |
| Performance Range / Specifications | Excellent thermostability: tolerates reaction temperatures up to 60 °C; retains approximately 70% residual activity after 30 min incubation at 58 °C. Suitable for RT-PCR and RT-qPCR. Synthesizes full-length cDNA up to 10 kb. A glycerol-free format is available upon request to meet different formulation needs. |
| Sensitivity / LOD | Sensitivity down to 1 pg total RNA; detection range 1 pg to 1 ug total RNA in RT-qPCR applications. |
| Reaction Conditions / Protocol | Recommended reaction temperature up to 60 °C (e.g., 50-60 °C) for reverse transcription of RNA templates with complex secondary structures. |
| Components / Formulation | Thermostable reverse transcriptase supplied as a liquid enzyme formulation; a glycerol-free version is available for lyophilization-compatible or special formulation requirements. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 2 years |
| Package Specifications | 10000 U; 5 x 10000 U; 200000 U |
| Product Form | Liquid enzyme solution (glycerol-free format available) |
| Quality Control | Residual activity verified after heat challenge (approximately 70% activity retained after 30 min at 58 °C). |
| Source / Origin | Recombinant enzyme genetically engineered from M-MLV (H-) (Moloney murine leukemia virus, RNase H-deficient) reverse transcriptase |
| Expiration Date / Stability | Stable for 2 years at -25 to -15 °C. |
| Compatibility | Compatible with RT-PCR and RT-qPCR workflows, including one-step RT-qPCR formulations. |
| Application Notes / Precautions | RNase H-deficient (derived from an M-MLV (H-) backbone); particularly recommended for low-abundance templates, low-copy genes, and RNA templates with complex secondary structures; higher reaction temperatures (up to 60 °C) help resolve stable RNA secondary structures. |
| Heat Inactivation | Highly thermostable: retains approximately 70% activity after 30 min incubation at 58 °C; supports reverse transcription at up to 60 °C. |
