Product Name Restriction Enzyme SspDI (KasI) (Recognition Site: 5'-G↓GCGCC-3')
Catalog No. IVDRM-0160
Description SspDI (KasI) is a conventional restriction endonuclease that recognizes the G/GCGCC sequence. Unlike rapid restriction enzymes, SspDI (KasI) requires a longer digestion time to achieve complete cleavage of DNA substrates, but it uses the same universal reaction buffer as the rapid-enzyme series, making it easy to combine with other restriction enzymes in double digestions.
Intended Use Site-specific cleavage of DNA at G/GCGCC recognition sites for molecular cloning and restriction analysis, including double digestion with other restriction enzymes.
Principle / Technology Type II restriction endonuclease that recognizes the palindromic sequence 5'-G↓GCGCC-3' and cleaves double-stranded DNA at defined positions within the site.
Sample Type Double-stranded DNA substrates such as pBR322.
Performance Range / Specifications Concentration: 10 U/μL
Requires extended digestion time for complete cleavage of DNA substrates
Isoschizomers: KasI, DinI, EgeI, EheI, Mly113I, NarI, PluTI, SfoI
Specificity Specifically recognizes and cleaves 5'-G↓GCGCC-3' / 3'-CCGCG↑G-5'.
Reaction Conditions / Protocol 1× universal reaction buffer; incubate at 37°C. Allow a longer digestion time for complete cleavage. Heat inactivation: 80°C for 20 min.
Components / Formulation SspDI (KasI) restriction enzyme (10 U/μL); universal reaction buffer.
Storage Conditions Store at -25 to -15°C.
Shelf Life 2 years when stored at -25 to -15°C.
Package Specifications 250 U
Concentration 10 U/μL
Activity / Unit Definition One unit (U) is defined as the amount of enzyme required to completely digest 1 μg of pBR322 DNA in a 50 μL reaction at 37°C within 1 h.
Expiration Date / Stability Stable for 2 years at -25 to -15°C.
Compatibility Isoschizomers: KasI, DinI, EgeI, EheI, Mly113I, NarI, PluTI, SfoI.
Recommended Buffer System 1× universal reaction buffer (shared with the rapid restriction enzyme series).
Application Notes / Precautions SspDI (KasI) is a conventional (non-rapid) enzyme; extend the digestion time to ensure complete cleavage.
Uses the universal reaction buffer, enabling double digestion with other restriction enzymes.
Recognition Site 5'-G↓GCGCC-3'
3'-CCGCG↑G-5'
Heat Inactivation 80°C for 20 min.
Methylation Sensitivity CpG: cleavage blocked.
Dam, Dcm, EcoKI, EcoBI: no effect.
