| Product Name | Restriction Enzyme SgrAI (Recognition Site: 5'-CR↓CCGGYG-3') |
| Catalog No. | IVDRM-0159 |
| Description | SgrAI is a restriction endonuclease originally derived from Streptomyces griseus, produced by recombinant expression in E. coli and purified for consistent performance. It specifically recognizes and cleaves the CR/CCGGYG sequence. SgrAI is supplied outside the rapid-enzyme series but is fully compatible with the universal reaction buffer, allowing convenient double digestion with other restriction enzymes. Owing to its distinctive mechanism, SgrAI achieves efficient cleavage only when two or more recognition sites are present, and its cleavage efficiency drops markedly on single-site substrates. |
| Intended Use | Site-specific cleavage of DNA at CR/CCGGYG recognition sites for molecular cloning, restriction analysis, and double-digestion workflows with other restriction enzymes. |
| Principle / Technology | Recombinant Type II restriction endonuclease that recognizes the degenerate sequence CR↓CCGGYG. Its efficient cleavage requires the presence of two or more recognition sites, reflecting a site-cooperative (allosteric) activation mechanism; single-site substrates are cleaved with significantly reduced efficiency. |
| Sample Type | Double-stranded DNA substrates such as λDNA. |
| Performance Range / Specifications | Concentration: 10 U/μL Efficient cleavage requires two or more recognition sites; significantly reduced efficiency on single-site substrates Compatible with the universal reaction buffer for double digestion with other restriction enzymes |
| Specificity | Specifically recognizes and cleaves 5'-CR↓CCGGYG-3' / 3'-GYGGCC↑RC-5'. |
| Reaction Conditions / Protocol | 1×Cut Buffer B; incubate at 37°C. Heat inactivation: 80°C for 20 min. For efficient cleavage, substrates should contain two or more SgrAI recognition sites. |
| Components / Formulation | SgrAI restriction enzyme (10 U/μL); 1×Cut Buffer B reaction buffer. |
| Storage Conditions | Store at -25 to -15°C. |
| Shelf Life | 2 years when stored at -25 to -15°C. |
| Package Specifications | 500 U |
| Concentration | 10 U/μL |
| Activity / Unit Definition | One unit (U) is defined as the amount of enzyme required to completely digest 1 μg of λDNA in a 50 μL reaction at 37°C within 1 h. |
| Source / Origin | Derived from Streptomyces griseus; recombinantly expressed in E. coli and purified. |
| Expiration Date / Stability | Stable for 2 years at -25 to -15°C. |
| Recommended Buffer System | 1×Cut Buffer B; compatible with the universal reaction buffer for double digestion with other restriction enzymes. |
| Application Notes / Precautions | Use substrates containing two or more recognition sites for efficient cleavage; single-site substrates are cleaved with markedly lower efficiency. Compatible with the universal reaction buffer, enabling double digests with other restriction enzymes. |
| Recognition Site | 5'-CR↓CCGGYG-3' 3'-GYGGCC↑RC-5' |
| Heat Inactivation | 80°C for 20 min. |
| Methylation Sensitivity | CpG, EcoKI, EcoBI: cleavage blocked. |
