Product Name Restriction Enzyme BsrDI (Recognition Site: 5'-GCAATGNN↓-3')
Catalog No. IVDRM-0155
Description BsrDI, derived from Bacillus stearothermophilus D70, is a Type IIS restriction enzyme that recognizes the sequence GCAATG/CATTGC. It cleaves the top strand two nucleotides downstream of the recognition site and the bottom strand immediately after the recognition site, generating specific cohesive ends, and can be used for Golden Gate assembly.
Intended Use Molecular cloning and Golden Gate assembly.
Principle / Technology As a Type IIS restriction enzyme, BsrDI recognizes the specific sequence GCAATG/CATTGC and cleaves outside the recognition site—two nucleotides downstream on the top strand and directly after the recognition site on the bottom strand—to produce specific cohesive (sticky) ends.
Sample Type DNA substrates (e.g., λ DNA).
Performance Range / Specifications Concentration: 10 U/μL.
Optimal reaction temperature: 37°C.
One unit (U) completely digests 1 µg of λ DNA in 1 h at 37°C in a 50 μL reaction.
Specificity Specifically recognizes 5'-GCAATG-3' and cleaves outside the recognition sequence to generate defined sticky ends.
Reaction Conditions / Protocol 1× BsrDI Buffer; incubate at 37°C.
Heat inactivation: 80°C for 20 min.
Components / Formulation BsrDI restriction enzyme (10 U/μL) with a dedicated 1× BsrDI reaction buffer.
Storage Conditions Store at -25 to -15°C.
Shelf Life 2 years.
