| Product Name | Restriction Enzyme BspQI (Recognition Site: 5'-GCTCTTC(N)1/(N)4-3') |
| Catalog No. | IVDRM-0222 |
| Description | This product is a Type II restriction endonuclease supplied as a recombinant protein expressed in E. coli from the BspQI gene of Bacillus sphaericus. It recognizes the sequence 5'-GCTCTTC(N)1/(N)4-3' and is used to digest plasmids to generate linearized DNA fragments with defined poly(A/T/G/C) ends and specific sticky ends. The product is supplied in liquid form. |
| Intended Use | Plasmid linearization for the preparation of in vitro transcription (IVT) templates, including mRNA vaccine development; generation of specific cohesive ends. As a Type IIS enzyme, BspQI cuts outside its recognition sequence, preserving the integrity of designed poly(A) tails and leaving no scar or unwanted nucleotides on the linearized DNA template. It is the most frequently used enzyme for plasmid linearization in mRNA development projects. |
| Principle / Technology | Type IIS restriction endonuclease activity: the enzyme binds the recognition sequence 5'-GCTCTTC(N)1/(N)4-3' and cleaves the DNA at a fixed distance outside the recognition site, producing defined sticky ends. |
| Performance Range / Specifications | High digestion efficiency with low star activity; no star activity observed after 16 h of reaction at 37 °C. Excellent application performance comparable to leading products. |
| Reaction Conditions / Protocol | Optimal reaction temperature: 50 °C. In a 50 uL reaction system, digest 1 ug of lambda DNA with an appropriate amount of BspQI at 50 °C for 60 min, then heat at 80 °C for 20 min to inactivate the enzyme. Star activity: none after 16 h at 37 °C. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 1 year |
| Package Specifications | 500 U 2500 U 10 KU 100 KU |
| Product Form | Liquid |
| Quality Control | Produced and quality-controlled under a strict quality management system; animal-origin-free (no animal-derived components introduced). |
| Concentration | 10 U/uL |
| Source / Origin | Recombinant protein expressed in E. coli, encoded by the BspQI gene derived from Bacillus sphaericus. |
| Regulatory / Compliance | Quality-controlled manufacturing under a certified quality management system; free of animal-derived components. |
| Compatibility | Suitable for linearization of plasmid templates bearing poly(A/T/G/C) tails; generates specific sticky ends for downstream applications. |
| Application Notes / Precautions | For IVT template preparation, select enzymes that generate 5' overhangs or blunt ends (3' overhangs increase aberrant by-products in IVT) and that have long recognition sequences to minimize cutting within the gene fragment. Type IIS enzymes are preferred for plasmid linearization in vaccine development because their cleavage sites lie outside the recognition sequence. |
| Recognition Site | 5'-GCTCTTC(N)1/(N)4-3' |
| Heat Inactivation | 80 °C for 20 min |
