| Product Name | Restriction Enzyme BspEI (Recognition Site: 5'-T↓CCGGA-3') |
| Catalog No. | IVDRM-0226 |
| Description | BspEI is a Type IIP restriction endonuclease originally derived from a Bacillus species and obtained by recombinant expression and purification from E. coli. It recognizes and cleaves the sequence 5'-TCCGGA-3', generating cohesive ends with a 4-base 5' overhang. BspEI is supplied with its dedicated Cut Buffer C and is not compatible with universal fast-digestion buffer. |
| Intended Use | For molecular cloning and other molecular biology research applications. For research use only. |
| Principle / Technology | Type IIP restriction endonuclease; sequence-specific double-stranded DNA cleavage producing a 4-base 5' protruding cohesive end. |
| Detection Method | Cleavage products are typically analyzed by agarose gel electrophoresis. |
| Sample Type | Double-stranded DNA (e.g., plasmid DNA, lambda DNA). |
| Specificity | Isoschizomers: Kpn2I, MroI, AccIII, Aor13HI, Bsp13I, BseAI. Cleaves 5'-TCCGGA-3' to generate a 4-base 5' overhang. |
| Reaction Conditions / Protocol | 1x Cut Buffer C; incubate at 37°C. |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | 2 years when stored as recommended. |
| Package Specifications | 500 U |
| Concentration | 10 U/uL |
| Activity / Unit Definition | One unit (U) is defined as the amount of enzyme required to completely digest 1 ug of lambda DNA (Dam-) in 1 hour at 37°C in a 50 uL reaction. |
| Source / Origin | Derived from a Bacillus species; recombinantly expressed and purified from E. coli. |
| Expiration Date / Stability | Stable for 2 years at -25°C to -15°C. |
| Compatibility | Uses the dedicated Cut Buffer C; not compatible with universal fast-digestion buffer. |
| Recommended Buffer System | 1x Cut Buffer C |
| Application Notes / Precautions | Do not substitute universal fast-digestion buffer for the dedicated Cut Buffer C, as this will impair enzyme performance. |
| Recognition Site | 5'-T↓CCGGA-3' 3'-AGGCC↑T-5' |
| Heat Inactivation | Incubate at 80°C for 20 min. |
| Methylation Sensitivity | Dam: cleavage blocked CpG: cleavage impaired Dcm, EcoKI, EcoBI: no effect |
For research use only, not for clinical use.
