Product Name Restriction Enzyme BsmBI (Recognition Site: 5'-CGTCTC(N)1↓-3')
Catalog No. IVDRM-0153
Description BsmBI is a Type IIS restriction endonuclease that recognizes a non-palindromic sequence and cleaves outside its recognition site, making it a workhorse for Golden Gate assembly. The enzyme is supplied with an optimized reaction buffer that maximizes BsmBI activity, and the buffer contains recombinant albumin to enhance the stability of the enzyme.
Intended Use Molecular cloning and Golden Gate assembly.
Principle / Technology As a Type IIS restriction enzyme, BsmBI recognizes the non-palindromic sequence CGTCTC and cuts the DNA at fixed positions outside the recognition sequence (1 nt downstream on the top strand and 5 nt downstream on the bottom strand), generating defined cohesive ends for seamless assembly.
Sample Type DNA substrates (e.g., plasmid DNA).
Performance Range / Specifications Concentration: 10 U/μL.
Optimal reaction temperature: 55°C.
Supplied with an optimized reaction buffer that maximizes BsmBI activity.
Specificity Specifically recognizes the non-palindromic sequence 5'-CGTCTC-3' and cleaves outside the recognition site.
Reaction Conditions / Protocol 1× BsmBI Buffer; incubate at 55°C.
Heat inactivation: 80°C for 20 min.
Components / Formulation BsmBI restriction enzyme (10 U/μL) with an optimized reaction buffer containing recombinant albumin for enhanced enzyme stability.
Storage Conditions Store at -25 to -15°C.
Shelf Life 2 years.
Package Specifications 1000 U; 300 U
Concentration 10 U/μL
Expiration Date / Stability Valid for 2 years when stored at -25 to -15°C.
Recommended Buffer System 1× BsmBI Buffer.
Application Notes / Precautions For double digestions together with other restriction enzymes, a version of this enzyme supplied in a universal buffer is recommended.
Temperature-sensitive alternatives require extra care during use and storage; this enzyme offers superior cleavage efficiency.
Recognition Site 5'-CGTCTC(N)1↓-3'
3'-GCAGAG(N)5↑-5'
Heat Inactivation 80°C for 20 min.
Methylation Sensitivity CpG: complete sequence overlap, cleavage blocked.
EcoBI: sequence may overlap, cleavage may be affected.
Dam, Dcm, EcoKI: no effect.

For research use only, not for clinical use.
