| Product Name | Restriction Enzyme AarI (Recognition Site: 5'-CACCTGC(N)4↓-3') |
| Catalog No. | IVDRM-0148 |
| Description | AarI is a Type IIS restriction endonuclease and a complete isoschizomer of PaqCI. It recognizes the asymmetric sequence CACCTGC and cuts at a defined distance downstream of the site, generating a 4-nucleotide 5' overhang. Unlike most Type IIS enzymes, AarI requires two or more recognition sites for efficient cleavage. Complete digestion is achieved with the activator supplied with the enzyme; cleavage still occurs without the activator but does not go to completion. |
| Intended Use | Molecular cloning and other applications requiring Type IIS restriction digestion, such as cleavage outside the recognition sequence. |
| Principle / Technology | AarI is a Type IIS restriction endonuclease that recognizes the asymmetric sequence 5'-CACCTGC-3' and cleaves the DNA at fixed positions downstream of the recognition site, producing 4-nucleotide 5' overhangs. Efficient cleavage requires the presence of two or more recognition sites, and full digestion is supported by the activator provided with the enzyme. |
| Sample Type | DNA substrates such as lambda DNA (λDNA) bearing AarI recognition sites. |
| Performance Range / Specifications | Concentration: 5 U/μL. Unit definition: One unit (U) is the amount of enzyme required to completely digest 1 µg of λDNA in 1 hour at 37°C in a 50 µL reaction in the presence of the activator. Cleavage produces a 4-nucleotide 5' overhang. CpG methylation of the recognition site affects cleavage. |
| Specificity | Specifically recognizes 5'-CACCTGC(N)4↓-3' (complementary strand 3'-GTGGACG(N)8↑-5') and cleaves downstream of the recognition sequence. |
| Reaction Conditions / Protocol | Optimal reaction: incubate at 37°C in 1× universal reaction buffer. Use the supplied activator to achieve complete digestion. Heat inactivation: 80°C for 20 min. |
| Components / Formulation | AarI restriction enzyme (5 U/μL), 1× universal reaction buffer, and a dedicated activator for complete digestion. |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | Valid for 2 years when stored at -25°C to -15°C. |
| Package Specifications | 100 U |
| Concentration | 5 U/μL |
| Activity / Unit Definition | One unit (U) is defined as the amount of enzyme required to completely digest 1 µg of λDNA in 1 hour at 37°C in a 50 µL reaction system in the presence of the activator. |
| Expiration Date / Stability | Stable for 2 years from the date of manufacture when stored at -25°C to -15°C. |
| Compatibility | Isoschizomer: PaqCI. |
| Recommended Buffer System | 1× universal reaction buffer (supplied), with dedicated activator for complete digestion. |
| Application Notes / Precautions | AarI requires two or more recognition sites for efficient cleavage. Always add the supplied activator when complete digestion is required; without the activator, cleavage is incomplete. Inactivate the reaction by incubation at 80°C for 20 min when downstream steps are sensitive to residual enzyme activity. |
| Recognition Site | 5'-CACCTGC(N)4↓-3' 3'-GTGGACG(N)8↑-5' |
| Heat Inactivation | 80°C for 20 min. |
| Methylation Sensitivity | CpG methylation: cleavage is affected. |
