Description A high-purity, calcium-dependent recombinant endoprotease that selectively cleaves peptide bonds after paired basic residues and is supplied as a stable lyophilized powder.
Intended Use Fusion-tag removal and processing of insulin analogs, GLP-1 products and recombinant peptides, including workflows that combine Kex2 protease with carboxypeptidase B.
Principle / Technology The enzyme recognizes dibasic sequences such as Arg-Arg and Lys-Arg and cleaves the peptide bond on the carboxyl side. It does not cleave after a single basic residue and is not inhibited by aprotinin, PMSF or TPCK.
Performance Range / Specifications Recommended reconstituted enzyme concentration: approximately 1-10 mg/mL.
Specificity Selective cleavage after paired basic amino acid residues; no cleavage after an isolated basic residue.
Reaction Conditions / Protocol Use 50 mM Tris-HCl with 2 mM Ca2+ at pH 7.0-9.0 as the reaction buffer. For storage after reconstitution, dissolve in 20 mM sodium acetate-acetic acid with 2 mM Ca2+ at pH 5.2, aliquot and freeze below -20°C.
