| Product Name | Recombinant DNase I Set (RNase-Free, Yeast-Expressed) |
| Catalog No. | IVDRM-0093 |
| Description | DNase I (Deoxyribonuclease I) is an endonuclease that digests single-stranded and double-stranded DNA. It hydrolyzes phosphodiester bonds, producing mono- and oligodeoxynucleotides with 5'-phosphate and 3'-OH groups. DNase I works optimally at pH 7-8; its activity depends on Ca2+ and can be activated by divalent metal ions such as Co2+, Mn2+, and Zn2+. In the presence of Mg2+, DNase I cleaves double-stranded DNA at random sites, whereas in the presence of Mn2+ it cuts both DNA strands at approximately the same site, generating blunt ends or sticky ends with 1-2 nucleotide overhangs. This recombinant, yeast-expressed DNase I set is RNase-free and delivers strong digestion capacity even in complex sample environments, making it suitable for processing a wide variety of RNA samples. |
| Intended Use | Digestion of single-stranded and double-stranded DNA, including genomic DNA removal during the processing of various RNA samples such as RNA extraction workflows. |
| Principle / Technology | DNase I is an endonuclease that hydrolyzes phosphodiester bonds in DNA, yielding mono- and oligodeoxynucleotides carrying 5'-phosphate and 3'-OH groups. Activity is Ca2+-dependent and can be activated by divalent metal ions (Co2+, Mn2+, Zn2+). With Mg2+, double-stranded DNA is cleaved at random sites; with Mn2+, both strands are cut at approximately the same site, producing blunt ends or sticky ends with 1-2 nucleotide overhangs. |
| Sample Type | Various RNA samples, including complex sample environments; validated on plasmid DNA digestion and mouse liver pre-treatment samples from 12 different sources for RNA extraction. |
| Performance Range / Specifications | Strong digestion capacity with efficient DNA digestion even in complex sample environments; plasmid DNA (1 μg) removal efficiency comparable to leading competing products. |
| Specificity | Digests single-stranded and double-stranded DNA without affecting RNA quality; no detectable RNase activity, eliminating the risk of RNA sample degradation. |
| Reaction Conditions / Protocol | Activity depends on Ca2+ and is activated by divalent metal ions such as Co2+, Mn2+, and Zn2+. In the presence of Mg2+, DNase I randomly cleaves double-stranded DNA; in the presence of Mn2+, it cuts both strands at approximately the same site. |
| Components / Formulation | Recombinant DNase I Set (RNase-free, yeast-expressed); set components as supplied per package size (50 T / 250 T). |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | 2 years when stored at -25°C to -15°C. |
| Package Specifications | 250 T, 50 T |
| Quality Control | No detectable RNase activity: 10 U of enzyme incubated with RNA substrate at 37°C for 1 h showed no RNase activity residue across three batches by agarose gel electrophoresis. Application validation: treatment of 12 mouse liver pre-treatment samples of different origins with 20 U of enzyme yielded RNA with good integrity after subsequent extraction. |
| Source / Origin | Recombinant, expressed in yeast. |
| pH Range / Optimal pH | Optimal working pH range: 7-8. |
| Expiration Date / Stability | Valid for 2 years at -25°C to -15°C. |
| Compatibility | Suitable for processing various RNA samples; digests DNA without compromising RNA integrity, meeting the technical requirements of RNA extraction experiments. |
| Application Notes / Precautions | DNase I activity is Ca2+-dependent and can be activated by divalent metal ions such as Co2+, Mn2+, and Zn2+. Choose the divalent ion according to the desired cleavage pattern: Mg2+ for random cleavage of double-stranded DNA; Mn2+ for cutting both strands at approximately the same site, producing blunt ends or sticky ends with 1-2 nucleotide overhangs. |
| Batch-to-Batch Consistency | Three batches of the enzyme preparation showed no detectable RNase activity residue when 10 U was incubated with RNA substrate at 37°C for 1 h, ensuring reliable protection for high-purity RNA analysis. |
