| Product Name | Proteinase K Solution (20 mg/mL, RNase-Free, DNase-Free) |
| Catalog No. | IVDRM-0188 |
| Description | Proteinase K is a broad-spectrum serine protease with a molecular weight of approximately 29.3 kDa. It cleaves peptide bonds at the carboxyl terminus of aliphatic and aromatic amino acids. Thanks to its robust and wide-ranging activity, Proteinase K is widely used for the preparation of chromosomal DNA for pulsed-field gel electrophoresis, for protein blotting workflows, and for eliminating nucleases from DNA and RNA preparations. Its activity is enhanced by denaturants such as 1% SDS, allowing efficient protein digestion even under challenging conditions. |
| Intended Use | Suitable for preparation of chromosomal DNA for pulsed-field gel electrophoresis, protein blotting applications, and removal of nucleases during DNA and RNA preparation. |
| Principle / Technology | Proteinase K is a serine protease that hydrolyzes peptide bonds at the carboxyl-terminal side of aliphatic and aromatic amino acids. Denaturants such as SDS (1%) can increase its activity. The enzyme is produced from a recombinant yeast strain. |
| Sample Type | DNA and RNA preparations, chromosomal DNA samples, and protein samples requiring proteolytic digestion or nuclease removal. |
| Performance Range / Specifications | Concentration: 20 mg/mL Molecular weight: approximately 29.3 kDa Activity is stable in the presence of urea and SDS Active over a broad pH range of 4.0-12.0, with an optimal pH range of 7.5-9.0 Free of RNase and DNase |
| Specificity | Cleaves peptide bonds at the carboxyl terminus of aliphatic and aromatic amino acids, providing broad proteolytic activity. |
| Reaction Conditions / Protocol | The typical working concentration is 50-100 µg/mL. The exact working concentration should be determined according to buffer composition, including the presence or absence of SDS and urea, as well as pH and temperature. The enzyme is active over pH 4.0-12.0, with optimal activity at pH 7.5-9.0, and its activity can be enhanced by 1% SDS. |
| Components / Formulation | Ready-to-use Proteinase K solution at 20 mg/mL. Derived from a recombinant yeast strain. |
| Storage Conditions | Store at 2-8°C. For longer-term storage, the product can be kept frozen at -25 to -15°C. |
| Shelf Life | 2 years when stored at 2-8°C. |
