| Product Name | Proteinase K Powder (RNase-Free, DNase-Free) |
| Catalog No. | IVDRM-0179 |
| Description | Proteinase K is a broad-spectrum serine protease with a molecular weight of approximately 29 kDa. It cleaves peptide bonds at the carboxyl side of aliphatic and aromatic amino acids, making it a versatile tool for molecular biology workflows. Typical applications include preparation of chromosomal DNA for pulsed-field gel electrophoresis, Western blotting, and removal of nucleases from DNA and RNA preparations. Denaturants such as 1% SDS enhance its activity. The commonly used working concentration is 50-100 µg/mL, and the optimal concentration should be determined according to buffer composition, including the presence of SDS or urea, pH, and temperature. |
| Intended Use | Digestion and removal of proteins during nucleic acid extraction and purification; sample preparation for in situ hybridization. |
| Principle / Technology | A serine protease that cleaves peptide bonds carboxyl-terminal to aliphatic and aromatic amino acids, efficiently degrading proteins and inactivating nucleases in nucleic acid samples. |
| Sample Type | DNA and RNA preparations; in situ hybridization samples; chromosomal DNA for pulsed-field electrophoresis. |
| Performance Range / Specifications | Retains activity in the presence of urea and SDS. Active over a broad pH range of 4.0-12.0, with an optimal pH range of 7.5-9.0. Typical working concentration: 50-100 µg/mL. |
| Specificity | Cleaves peptide bonds at the carboxyl side of aliphatic and aromatic amino acids. Certified free of RNase and DNase activity. |
| Reaction Conditions / Protocol | Commonly used at 50-100 µg/mL. The exact working concentration should be optimized according to whether the buffer contains SDS or urea, as well as the pH and temperature. Denaturants such as 1% SDS enhance enzymatic activity. |
| Components / Formulation | Lyophilized Proteinase K powder, RNase-free and DNase-free. |
| Storage Conditions | Store dry at 2-8°C. For longer-term storage, keep frozen at -25 to -15°C. |
| Shelf Life | 3 years when stored at 2-8°C. |
| Package Specifications | 100 mg; 1 g; 5 g; 10 g; 100 g |
| Product Form | Lyophilized powder |
| Quality Control | Tested free of RNase and DNase. Activity remains stable in the presence of urea and SDS. |
| Molecular Weight | Approximately 29 kDa |
| Source / Origin | Recombinant yeast strain |
| pH Range / Optimal pH | Active from pH 4.0 to 12.0; optimal pH 7.5-9.0. |
| Expiration Date / Stability | Stable for 3 years at 2-8°C; for extended storage, keep at -25 to -15°C. |
| Compatibility | Activity is stable in the presence of urea and SDS; denaturants such as 1% SDS enhance activity. |
| Application Notes / Precautions | Main applications include in situ hybridization sample preparation and digestion/removal of proteins during nucleic acid purification. Adjust the working concentration according to buffer composition (SDS, urea), pH, and temperature. |
