| Product Name | Murine RNase Inhibitor (40 U/μL) |
| Catalog No. | IVDRM-0249 |
| Description | Murine RNase Inhibitor is a recombinant ribonuclease inhibitor expressed in soluble form in E. coli. It inactivates RNases by forming a tight non-covalent complex with the enzyme, and broadly inhibits RNase A, RNase B and RNase C. Compared with the human placental inhibitor, it lacks the two oxidation-sensitive cysteine residues of the human protein, providing higher resistance to oxidation and making it better suited to DTT-sensitive applications such as qPCR. It has been verified by RT-PCR and RT-qPCR to be compatible with a wide range of reverse transcriptases and DNA polymerases, and is ideal for first-strand cDNA synthesis, polysome isolation, in vitro transcription and in vitro cell-free translation systems. |
| Intended Use | Supplied as a raw material for molecular diagnostic and life-science reagent manufacturing. Typical applications: First-strand cDNA synthesis Polysome isolation In vitro transcription In vitro cell-free translation systems RNA protection in RT-PCR and RT-qPCR workflows |
| Principle / Technology | The inhibitor binds RNase non-covalently to form a stable complex, resulting in specific and reversible inactivation of the RNase. |
| Performance Range / Specifications | Broad-spectrum RNase inhibition: inhibits RNase A, RNase B and RNase C Wide reaction compatibility: active from pH 5.0 to 9.0 and from 25 °C to 60 °C; suitable for use with thermostable reverse transcriptases at 55-60 °C No effect on the activity of SP6, T7 or T3 RNA polymerases, AMV or M-MLV reverse transcriptases, or Taq DNA polymerase Large-scale production: single-batch capacity of up to 2 billion units ensures lot uniformity, stability and reliable supply |
| Specificity | Inhibits RNase A, RNase B and RNase C. Does not inhibit or interfere with SP6, T7 or T3 RNA polymerases, AMV or M-MLV reverse transcriptases, or Taq DNA polymerase. |
| Reaction Conditions / Protocol | Active over pH 5.0-9.0 and 25-60 °C; compatible with thermostable reverse transcriptases operating at 55-60 °C. |
| Components / Formulation | Storage buffer: 20 mM HEPES-KOH, 150 mM KCl, 8 mM DTT, 0.5% NP-40, 0.5% Tween-20, 50% glycerol, pH 7.5 ± 0.2 at 25 °C. |
| Storage Conditions | Store at -25 °C to -15 °C. |
| Shelf Life | 2 years when stored at -25 °C to -15 °C. |
| Package Specifications | 2 KU 10 KU 20 KU 100 KU 1,000 KU 20,000 KU |
| Product Form | Liquid (supplied in storage buffer containing 50% glycerol) |
| Quality Control | Free of exonuclease contamination: 200 U of enzyme incubated with 0.5 μg λDNA-HindIII digest in a 20 uL reaction at 37 °C for 4 hours shows no degradation. Free of nicking endonuclease contamination: 200 U of enzyme incubated with 0.5 μg plasmid DNA in a 20 uL reaction at 37 °C for 4 hours shows no nicking activity. Free of RNase contamination: 200 U of enzyme incubated with 0.5 μg 293T total RNA in a 20 uL reaction at 37 °C for 4 hours shows no change in RNA banding by agarose gel electrophoresis. Each lot is tested against these specifications (data shown for three independent lots). |
| Concentration | 40 U/uL |
| Source / Origin | Recombinant murine RNase inhibitor gene expressed in soluble form in E. coli and purified. |
| pH Range / Optimal pH | Active over pH 5.0-9.0 |
| Expiration Date / Stability | Stable for 2 years at -25 °C to -15 °C. |
| Regulatory / Compliance | Manufactured under a certified quality management system with quality control testing performed against defined quality standards. |
| Compatibility | Compatible with a broad range of reverse transcriptases and DNA polymerases, as verified by RT-PCR and RT-qPCR. Does not affect SP6, T7 or T3 RNA polymerases, AMV or M-MLV reverse transcriptases, or Taq DNA polymerase. |
| Recommended Buffer System | Supplied in 20 mM HEPES-KOH, 150 mM KCl, 8 mM DTT, 0.5% NP-40, 0.5% Tween-20, 50% glycerol, pH 7.5 ± 0.2 at 25 °C. |
| Application Notes / Precautions | Lacks the two oxidation-sensitive cysteines present in the human placental inhibitor, providing higher antioxidant activity; well suited to high-DTT-sensitive applications such as qPCR. |
| Batch-to-Batch Consistency | Mature protein expression and purification platform with quality control testing against defined standards ensures batch-to-batch consistency; single-batch production capacity of up to 2 billion units supports lot uniformity. |
