Product Name Hot-Start Super Specific Taq DNA Polymerase (5 U/μL, for Genotyping)
Catalog No. IVDRM-0265
Description This hot-start Taq DNA polymerase is blocked with a proprietary antibody that suppresses the 5'→3' polymerase activity during reaction setup and ramp-up, preventing non-specific amplification. The blocking antibody is fully inactivated by a 30-second heating step at the pre-denaturation temperature, releasing full DNA polymerase activity. The enzyme delivers high amplification efficiency, specificity, and sensitivity, and is well suited for Amplification Refractory Mutation System (ARMS) PCR and other high-specificity PCR applications.
Intended Use Designed for SNP genotyping, ARMS-PCR, and other high-specificity PCR amplification in molecular diagnostic assay development.
Principle / Technology Antibody-mediated hot-start technology: a specific antibody blocks the 5'→3' polymerase activity of Taq DNA polymerase at low temperatures. A 30-second incubation at the pre-denaturation temperature irreversibly inactivates the antibody and restores full polymerase activity.
Detection Method PCR / ARMS-PCR (endpoint or real-time amplification-based detection)
Sample Type Purified DNA templates, including mixed samples containing low proportions of mutant allele against a wild-type background
Performance Range / Specifications Strong specificity for SNP genotyping
In an ARMS-PCR premix, reliably detects 0.05% mutant template while showing no amplification of 100% wild-type template
Sensitivity / LOD Detects mutant template present at 0.05% against a wild-type background in ARMS-PCR
Specificity High; no amplification observed with 100% wild-type template in ARMS-PCR genotyping assays
Reaction Conditions / Protocol Hot-start activation: heat at the pre-denaturation temperature for 30 sec to fully inactivate the blocking antibody and release polymerase activity; then proceed with standard PCR cycling.
