| Product Name | Hot-Start J-Taq DNA Polymerase (6 U/μL, Glycerol-Free) |
| Catalog No. | IVDRM-0259 |
| Description | Hot-Start J-Taq DNA Polymerase (6 U/μL, Glycerol-Free) is a hot-start DNA polymerase in which enzyme activity is blocked by a specific antibody, delivering excellent amplification sensitivity and specificity. The blocking antibody is fully denatured within 30 sec at the pre-denaturation temperature, releasing full DNA polymerase activity. This hot-start mechanism effectively suppresses amplification caused by nonspecific primer annealing. The formulation is completely glycerol-free and is specifically designed for the preparation of lyophilized (freeze-dried) reagents. |
| Intended Use | Raw material enzyme for PCR-based molecular assays, especially for the formulation of lyophilized PCR reagents; suitable for applications such as methylation detection, bisulfite-tolerant amplification, and amplification of uracil-containing templates. |
| Principle / Technology | Antibody-mediated hot-start technology: a thermolabile blocking antibody binds and inactivates the polymerase at lower temperatures, preventing nonspecific primer annealing and extension during reaction setup. Upon heating at the pre-denaturation temperature for 30 sec, the antibody is irreversibly denatured and full polymerase activity is restored. |
| Sample Type | DNA templates, including bisulfite-treated DNA and uracil-containing DNA templates |
| Performance Range / Specifications | Glycerol-free formulation suitable for preparing lyophilized reagents. High amplification sensitivity and specificity. Fast antibody release: full activity restored within 30 sec at pre-denaturation temperature. |
| Specificity | Antibody-based hot start effectively inhibits nonspecific amplification resulting from nonspecific primer annealing. |
| Reaction Conditions / Protocol | Activate the enzyme during the pre-denaturation step; the blocking antibody is fully inactivated after 30 sec at the pre-denaturation temperature. |
| Components / Formulation | Glycerol-free hot-start DNA polymerase solution at 6 U/μL. |
| Storage Conditions | Store at 2-8°C. |
| Shelf Life | 2 years when stored at 2-8°C. |
| Package Specifications | 90 KU 600 U 15 KU |
| Product Form | Glycerol-free liquid formulation, 6 U/μL; designed for lyophilized reagent preparation. |
| Concentration | 6 U/μL |
| Expiration Date / Stability | Valid for 2 years when stored at 2-8°C. |
| Compatibility | Broad compatibility for PCR-based assay development, including lyophilized reagent systems. |
| Application Notes / Precautions | This product contains no glycerol and is specifically intended for formulating lyophilized (freeze-dried) reagents. A 30 sec pre-denaturation step is sufficient to fully release polymerase activity. |
