| Product Name | Hot-Start J-Taq DNA Polymerase (5 U/μL, for Multiplex PCR) |
| Catalog No. | IVDRM-0240 |
| Description | This product is an antibody-blocked, thermostable mutant Taq DNA polymerase. Its polymerase activity remains inhibited at 60 °C and is released only after heating at 95 °C, thereby preventing non-specific amplification and primer-dimer formation during sample preparation and reaction ramp-up. Like wild-type Taq DNA polymerase, J-Taq possesses 5'->3' polymerase activity and 5'->3' exonuclease activity, but lacks 3'->5' exonuclease (proofreading) activity. |
| Intended Use | Raw material enzyme for high-sensitivity PCR and multiplex PCR assay development in molecular diagnostics. |
| Principle / Technology | Antibody-based hot-start technology: a blocking antibody suppresses polymerase activity at ambient temperature and up to 60 °C; activity is released by 95 °C heating during initial denaturation. |
| Detection Method | PCR / real-time PCR |
| Performance Range / Specifications | High specificity: antibody blocking suppresses polymerase activity at room temperature, improving reaction specificity. No nuclease residue: free of exonuclease, endonuclease, and RNase contamination. Suitable for high-sensitivity PCR and multiplex PCR experiments. |
| Specificity | Antibody blocking suppresses polymerase activity at ambient temperature, effectively preventing non-specific amplification and primer-dimers during reaction setup and ramp-up. |
| Reaction Conditions / Protocol | Hot-start activation: polymerase activity remains inhibited at 60 °C and is released after heating at 95 °C (initial denaturation step). |
| Components / Formulation | Antibody-blocked mutant Taq DNA polymerase supplied at 5 U/uL. |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 2 years |
| Package Specifications | 250 U; 500 U; 1000 U; 10000 U; 25000 U |
| Product Form | Liquid enzyme solution, 5 U/uL |
| Quality Control | Tested free of exonuclease, endonuclease, and RNase residue. |
| Purity | No detectable exonuclease, endonuclease, or RNase residue. |
| Concentration | 5 U/uL |
| Source / Origin | Recombinant thermostable mutant Taq DNA polymerase expressed in E. coli |
| Expiration Date / Stability | Stable for 2 years at -25 to -15 °C. |
| Compatibility | Suitable for high-sensitivity PCR and multiplex PCR applications. |
| Application Notes / Precautions | Possesses 5'->3' polymerase and 5'->3' exonuclease activities; lacks 3'->5' exonuclease (proofreading) activity, as with wild-type Taq DNA polymerase. |
| Heat Inactivation | Not applicable (thermostable enzyme); the hot-start blocking antibody is inactivated by heating at 95 °C to release polymerase activity. |
