| Product Name | Hot-Start J-Taq DNA Polymerase (40 U/μL, Glycerol-Free) |
| Catalog No. | IVDRM-0267 |
| Description | This hot-start DNA polymerase is supplied as a high-concentration, glycerol-free formulation. It is blocked with a proprietary antibody and delivers good amplification sensitivity and specificity. A 30-second heating step at the pre-denaturation temperature fully inactivates the blocking antibody and releases DNA polymerase activity. The hot-start mechanism effectively suppresses amplification caused by non-specific primer annealing. The glycerol-free, high-concentration format is designed for the preparation of lyophilized PCR systems, reducing cost and operational complexity. |
| Intended Use | For the development and manufacture of lyophilized (freeze-dried) PCR/qPCR reagents, including in-situ lyophilization and lyophilized microbead formats; also suitable for conventional liquid PCR systems. |
| Principle / Technology | Antibody-mediated hot-start technology: the blocking antibody suppresses Taq DNA polymerase activity at low temperatures and is fully inactivated by a 30-second incubation at the pre-denaturation temperature, releasing polymerase activity. |
| Detection Method | PCR / real-time PCR (qPCR) |
| Sample Type | Purified DNA templates |
| Performance Range / Specifications | Suitable for the preparation of lyophilized systems, reducing cost and operational complexity Supports customized lyophilization formulations Reference lyophilization process available to support freeze-drying workflow development Post-lyophilization performance is consistent with the liquid reagent, with full retention of enzyme activity Stable at room temperature after lyophilization, enabling ambient shipping and storage |
| Sensitivity / LOD | Good amplification sensitivity and specificity |
| Specificity | Antibody hot-start blocking effectively suppresses amplification caused by non-specific primer annealing |
| Reaction Conditions / Protocol | Hot-start activation: heat at the pre-denaturation temperature for 30 sec to fully inactivate the blocking antibody; then proceed with standard PCR cycling. |
