Product Name Hot-Start High-Tolerant Taq DNA Polymerase, Ultra-Clean (20 U/μL)
Catalog No. IVDRM-0268
Description This hot-start DNA polymerase is dual-blocked with two proprietary antibodies that suppress both the 5'→3' polymerase activity and the 5'→3' exonuclease activity of Taq DNA polymerase. It is an optimal DNA polymerase for PCR amplification of uracil-containing template DNA after bisulfite conversion. A 30-second heating step at the pre-denaturation temperature fully inactivates the blocking antibodies, releasing both polymerase and exonuclease activities. The dual-blocking design prevents non-specific amplification caused by mispriming or primer dimers and suppresses fluorescence signal decay caused by probe degradation, improving the stability of in vitro diagnostic reagents during shipping and room-temperature handling. Manufactured to an ultra-clean, low nucleic acid background standard, with residual E. coli genomic DNA controlled to ≤1 copy/μL.
Intended Use For methylation detection assays and melt curve analysis; optimal for PCR amplification of bisulfite-treated, uracil-containing DNA templates in epigenetic diagnostic applications.
Principle / Technology Dual-antibody hot-start technology: two blocking antibodies suppress the 5'→3' polymerase and 5'→3' exonuclease activities at low temperatures; both activities are restored after a 30-second incubation at the pre-denaturation temperature.
Detection Method PCR / real-time PCR (qPCR), including methylation-specific PCR and melt curve analysis
Sample Type Bisulfite-converted DNA templates containing uracil; low-methylation-level samples
Performance Range / Specifications Suitable for methylation detection projects; tolerates bisulfite-treated templates
Ultra-clean, low nucleic acid background: residual E. coli genomic DNA ≤1 copy/μL
In PCR premix testing of a commercial template with 0.5% methylation rate, showed better amplification than comparable products, with improved curve peak shape and detection advantage at low template concentrations
Sensitivity / LOD Detects templates with 0.5% methylation rate; advantageous amplification of low-concentration templates with better peak shape
Specificity Dual antibody blocking prevents non-specific amplification from mispriming and primer dimers and suppresses probe degradation-related fluorescence signal loss
Reaction Conditions / Protocol Hot-start activation: heat at the pre-denaturation temperature for 30 sec to fully inactivate the blocking antibodies; then proceed with standard PCR cycling.
