| Product Name | Hot-Start High-Specificity Taq DNA Polymerase (5 U/μL, for STR Genotyping) |
| Catalog No. | IVDRM-0263 |
| Description | This product is a blend of a high-affinity anti-Taq monoclonal antibody and a thermostable recombinant Taq DNA polymerase. The antibody binds Taq DNA polymerase with very high affinity and maintains complete blocking of polymerase activity even after incubation at 50 °C for 30 min; upon heating at the pre-denaturation temperature for 30 sec the antibody is fully inactivated, releasing DNA polymerase activity. Use of this hot-start Taq enzyme effectively suppresses amplification caused by non-specific primer annealing. The recombinant Taq DNA polymerase exhibits high template affinity and is highly suitable for amplification of low-copy-number templates. Additional stabilizing additives that enhance the blocking effect are included, further improving the room-temperature stability of the system. |
| Intended Use | STR genotyping and forensic DNA profiling; hot-start PCR and qPCR; amplification of low-copy-number templates; high-throughput sequencing library preparation. |
| Principle / Technology | Antibody-mediated hot start. A high-affinity anti-Taq monoclonal antibody blocks the polymerase activity of the thermostable recombinant Taq DNA polymerase at ambient and moderate temperatures, preventing non-specific amplification from mis-priming and primer dimers. During the initial pre-denaturation step, heating for 30 sec completely inactivates the antibody and releases active DNA polymerase. |
| Sample Type | DNA templates, including low-copy-number DNA and forensic/STR profiling samples |
| Performance Range / Specifications | Excellent stability: blocking-enhancing additives provide good room-temperature stability of the system High amplification efficiency: thermostable recombinant DNA polymerase with high template affinity, ideal for low-copy-number template amplification Broad applicability: suitable for conventional PCR, hot-start PCR, qPCR and high-throughput sequencing Blocking maintained after 50 °C for 30 min; full release of activity within 30 sec at pre-denaturation temperature |
| Sensitivity / LOD | High template affinity supports reliable amplification of low-copy-number templates |
| Specificity | Antibody-mediated hot start effectively suppresses non-specific amplification caused by non-specific primer annealing; no non-specific signal is observed in no-template controls (NTC) |
| Storage Conditions | Store at -25 to -15 °C |
| Shelf Life | 2 years at -25 to -15 °C |
| Package Specifications | 500 U; 5,000 U |
| Product Form | Liquid (enzyme-antibody complex with stabilizing additives) |
| Concentration | 5 U/uL |
| Compatibility | Compatible with conventional PCR, hot-start PCR, qPCR and high-throughput sequencing workflows |
| Application Notes / Precautions | Well suited for STR genotyping assays: delivers higher overall peak heights and stronger detection capability for positive controls in forensic kits, with no non-specific peaks in negative controls Recommended pre-denaturation step of 30 sec to fully activate the enzyme |
