Product Name High-Purity Deoxyribonuclease I for DNA Degradation, >=95% by SDS-PAGE
Catalog No. IEC-HMM-0179
Description Alta DiagnoTech deoxyribonuclease I is a high-purity 35 kDa glycoprotein nuclease supplied as white amorphous powder.
Intended Use Enzymatic degradation of DNA and DNase activity measurement using a calf-thymus DNA substrate.
Principle / Technology DNase I converts DNA into oligonucleotides, producing an activity-proportional increase in absorbance at 260 nm.
Detection Method Kunitz absorbance assay at 260 nm
Performance Range / Specifications Specific activity is at least 20,000 Kunitz units/mg protein; assay linear range is 400-500 U/mL.
Reaction Conditions / Protocol A 3.00 mL assay contains 83 mM sodium acetate at pH 5.0, 4.2 mM MgSO4, 25 mM NaCl, 33.33 ug/mL calf-thymus DNA and 200-250 Kunitz units of DNase I. Preincubate 2.5 mL reagent for 1.5 minutes at 25 degrees C, add 0.5 mL sample and record the maximum absorbance-change rate within 2 minutes.
Storage Conditions Store at -20 degrees C
Shelf Life At least 2 years at -20 degrees C
Product Form White amorphous powder
Purity >=95% by SDS-PAGE; free of RNase, protease and other DNA endonuclease or exonuclease activity
Activity / Unit Definition One Kunitz unit increases A260 by 0.001 per minute at pH 5.0 and 25 degrees C using calf-thymus DNA.
Molecular Weight Approximately 35 kDa glycoprotein
pH Range / Optimal pH Stable at pH 4.0-9.0 for 24 hours at 25 degrees C; optimum pH 6.0
Appearance White amorphous powder
Alias Deoxyribonuclease I
Product Type Deoxyribonuclease I enzyme raw material
Function / Efficacy Hydrolyze DNA into oligonucleotides.
Abbreviation DNase I
Customization Specifications can be customized
Specific Activity >=20,000 Kunitz units/mg protein
Isoelectric Point Approximately 6.0
Inhibitors EDTA, EGTA, SDS, DTT, beta-mercaptoethanol and other reducing agents
Optimal Temperature Activity increases from 20 to 60 degrees C; the value at 37 degrees C is 3.3 times the value at 25 degrees C
Thermal Stability Stable below 55 degrees C; retains more than 50% activity after 30 minutes at 60 degrees C; inactive above 70 degrees C
Working Solution Dilute the reconstituted enzyme to 200-500 Kunitz units/mL in 0.85% NaCl before activity measurement.
Activator Mg2+, Mn2+, Ca2+, Co2+ and Zn2+
Detection Wavelength 260 nm
