Product Name GMP-Grade Recombinant Enterokinase with ≤10 EU/kU Endotoxin
Catalog No. IEC-HMM-0210
Description Alta DiagnoTech GMP-grade recombinant enterokinase is an engineered, high-specificity DDDDK-cleaving protease for peptide-manufacturing workflows.
Intended Use For removal of affinity tags from N-terminal or Met-N-terminal fusion proteins and preparation of peptide products such as GLP-1 analogues.
Principle / Technology An engineered enterokinase light-chain fragment specifically cleaves after lysine in DDDDK sequences when the next residue is not proline.
Reaction Conditions / Protocol For 0.1-1 mg/mL fusion protein and 0.1-1 mg total protein, use 0.1-1 U enzyme and incubate at 25°C for 12-16 hours or overnight.
Storage Conditions Store at -25°C to -15°C.
Package Specifications 2 kU, 20 kU, 200 kU, 2 MU and 20 MU
Product Form Liquid enzyme
Quality Control Endotoxin ≤10 EU/kU; host-cell protein ≤0.01%; host-cell nucleic acid ≤2 ng/kU.
Purity ≥95% by SDS-PAGE; ≥90% by HPLC
Activity / Unit Definition One unit cleaves 95% of 0.5 mg fusion protein in 25 mM Tris-HCl, pH 8.0, within 12-16 hours at 25°C.
Source / Origin Recombinant Escherichia coli expression; engineered light-chain fragment with a 6×His tag
Shipping Conditions Ship on dry ice.
Recommended Buffer System 25 mM Tris-HCl, pH 8.0
Application Notes / Precautions Cleavage may be reduced above 200 mM imidazole, above 200 mM NaCl or above 5% glycerol. The 6×His-tagged enzyme may be removed by nickel affinity or DEAE ion exchange.
Appearance Clear, colorless to pale-yellow liquid
Alias GMP-Grade Recombinant Enterokinase
Product Type GMP-grade recombinant enterokinase
