Product Name Fast Restriction Enzyme SmaI (Recognition Site: CCC/GGG)
Catalog No. IVDRM-0079
Description A recombinant fast restriction enzyme engineered for rapid, precise DNA cleavage within 5–15 minutes. It is well suited for digesting plasmid DNA, PCR products, and genomic DNA. All enzymes in this series share a single universal reaction buffer, which greatly simplifies digestion setup, and exhibit generous activity headroom to handle excess substrate or difficult templates with ease.
Intended Use Intended for rapid restriction digestion of plasmid DNA, PCR products, and genomic DNA in molecular cloning and other molecular biology research applications.
Principle / Technology Recombinant fast restriction endonuclease produced by genetic engineering. It specifically recognizes the sequence CCC/GGG and cleaves double-stranded DNA at defined positions within 5–15 minutes. All enzymes in this series share one universal digestion buffer, simplifying reaction setup, and offer ample activity redundancy for challenging templates or excess substrate.
Sample Type Plasmid DNA, PCR products, genomic DNA
Performance Range / Specifications Complete, precise DNA digestion within 5–15 minutes; enzyme concentration 20 U/μL; shared universal digestion buffer across the series; high activity redundancy for excess substrate or difficult templates.
Specificity Highly sequence-specific cleavage:
5'-CCC↓GGG-3'
3'-GGG↑CCC-5'
Reaction Conditions / Protocol Recommended reaction conditions:
1× universal fast digestion buffer
Optimal incubation temperature: 25°C
Typical digestion time: 5–15 min
Storage Conditions Store at -25°C to -15°C.
Shelf Life 2 years when stored at -25°C to -15°C.
Package Specifications 100 T
Concentration 20 U/μL
Activity / Unit Definition Enzyme activity: 20 U/μL.
Source / Origin Recombinant enzyme produced by genetic engineering (recombinant expression).
Expiration Date / Stability Stable for 2 years when stored at -25°C to -15°C.
Recommended Buffer System A single universal fast digestion buffer is shared across the entire fast restriction enzyme series, simplifying reaction setup and enabling convenient double or multiple digests.
Application Notes / Precautions SmaI generates blunt ends upon cleavage.
The optimal incubation temperature is 25°C, not 37°C.
CpG methylation completely overlaps the recognition site and blocks cleavage.
For complete digestion of excess substrate or difficult templates, extend the incubation time or increase the enzyme amount appropriately.
Recognition Site 5'-CCC↓GGG-3'
3'-GGG↑CCC-5'
Heat Inactivation Incubate at 80°C for 20 min.
Methylation Sensitivity CpG methylation: the CpG site completely overlaps the recognition sequence; cleavage is blocked.

For research use only, not for clinical use.
