Product Name Double-Block Anti-Taq DNA Polymerase Monoclonal Antibody
Catalog No. IVDRM-0284
Description This double-block monoclonal antibody is designed for hot-start PCR. Upon binding to Taq DNA polymerase, it simultaneously inhibits the 5' to 3' polymerase activity and the 5' to 3' exonuclease activity of the enzyme. It effectively suppresses nonspecific primer annealing and primer-dimer-driven nonspecific amplification at low temperatures and effectively prevents probe degradation. The antibody is denatured during the initial DNA denaturation step of PCR, restoring DNA polymerase activity and achieving a hot-start effect. No special antibody inactivation treatment is required, and the product can be used under conventional PCR reaction conditions.
Intended Use Raw material for hot-start PCR/qPCR reagent formulation; used to simultaneously block the polymerase and 5' to 3' exonuclease activities of Taq DNA polymerase for high-specificity, high-sensitivity amplification and probe protection in molecular diagnostic assays.
Principle / Technology Dual antibody-mediated hot-start mechanism: the double-block monoclonal antibody binds Taq DNA polymerase and blocks both its 5' to 3' polymerase activity and 5' to 3' exonuclease activity at low temperature. During the initial DNA denaturation step of PCR (95 °C), the antibody is heat-denatured and dissociates, releasing and restoring enzyme activity for specific amplification.
Detection Method Hot-start PCR / qPCR (including ARMS-PCR and probe-based assays)
Performance Range / Specifications Superior amplification specificity: dual blocking of polymerase and exonuclease activities effectively avoids nonspecific amplification.
Excellent detection sensitivity: the hot-start format ensures effective detection of low-abundance genes.
Outstanding stability: performance remains stable after 5 days at 37 °C or 10 days at 4 °C.
Flat baseline with good linearity: resolves baseline drift issues.
Fast enzyme activity release: more than 95% of enzyme activity is released after heating at 95 °C for 20 s.
Broad enzyme compatibility: suitable for both wild-type and mutant Taq enzymes; each mg of antibody blocks 4-10 KU of Taq enzyme.
Sensitivity / LOD The hot-start format ensures effective detection of low-abundance genes with higher sensitivity; validated by accurate genotyping of positive samples in ARMS-PCR amplification.
Specificity Dual blocking of polymerase and exonuclease activities effectively avoids nonspecific amplification and improves amplification specificity; verified for wild-type and mutant Taq enzymes at a blocking ratio of 1 µg antibody to 5 U Taq enzyme.
Reaction Conditions / Protocol Recommended blocking ratio: 1 µg of antibody per 5 U of Taq DNA polymerase; each mg of antibody blocks 4-10 KU of Taq enzyme. No special antibody inactivation treatment is required; use under conventional PCR reaction conditions.
