Principle |
Cholesterol in blood primarily exists in the form of cholesterol esters. During the assay, cholesterol esterase hydrolyzes cholesterol esters into cholesterol. Cholesterol is then oxidized by cholesterol oxidase to produce hydrogen peroxide. Hydrogen peroxide is subsequently processed by enzymes and reacts with a probe to generate a red color (λmax = 570 nm) and fluorescence (Ex/Em = 535/587 nm). |

Procedure |
1. Measure cholesterol using the entire sample; or separate HDL and LDL/VLDL by mixing the sample with precipitation buffer, incubating for 10 minutes, centrifuging at 2,000 g for 10 minutes, retaining the supernatant of the HDL fraction, and repeating centrifugation and resuspending the LDL/VLDL fraction precipitate.
2. During analysis, add the sample and standard to the wells.
3. Add the total cholesterol reaction mixture (containing esterase) or free cholesterol reaction mixture (without esterase), and incubate at 37°C for 60 minutes.
4. Analyze using a microplate reader. |
