| Principle | This assay is based on the ability of oxygen to quench the excited state of an oxygen-sensitive probe. The probe is taken up by non-specific energy-dependent endocytosis, and after washing, cells are monitored on a fluorescence microplate reader (dual-reading TR-F is required for complete oxygen quantification). The probe's phosphorescence is quenched by intracellular oxygen in a non-chemical, reversible manner, enabling measurement of average intracellular O₂ levels and facilitating real-time monitoring of relative changes in cellular oxygen consumption. The probe signal intensifies as intracellular oxygen decreases and weakens as intracellular oxygen increases. The probe can be excited at 360–400 nm or 535 nm, with an emission wavelength of 630–680 nm. The optimal filter combination is Ex/Em = 340/642 nm. |
