| Product Name | Chicken-Heart L-Lactate Dehydrogenase with Dual 340 nm Activity Assays |
| Catalog No. | IEC-HMM-0230 |
| Description | Alta DiagnoTech L-lactate dehydrogenase is a white amorphous enzyme powder characterized by complementary lactate- and pyruvate-substrate activity methods. |
| Intended Use | Enzyme raw material and activity standard for lactate-dehydrogenase assay development. |
| Principle / Technology | In the lactate method, L-LDH oxidizes L-lactate with NAD+ to pyruvate and NADH; in the pyruvate method, it reduces pyruvate with NADH to L-lactate and NAD+. |
| Detection Method | Kinetic absorbance assay at 340 nm |
| Performance Range / Specifications | Specific activity is at least 70 U/mg dry powder by the lactate-substrate method and at least 100 U/mg dry powder by the pyruvate-substrate method. Recommended assay ranges are 200-2,000 U/L and 500-3,000 U/L, respectively. |
| Reaction Conditions / Protocol | For either method, preincubate 1,200 uL working reagent for 1 minute at 37 degrees C, add 20 uL enzyme sample and record the A340 change during minutes 2-3. Lactate-method reagent uses Buffer:R1:R2 at 920:80:200; pyruvate-method reagent uses Buffer:R1:R2 at 3,000:100:50. |
| Storage Conditions | Store dry powder at -20 degrees C |
| Shelf Life | At least 1 year at -20 degrees C |
| Product Form | White amorphous powder |
| Activity / Unit Definition | One unit generates 1 umol NADH per minute in the lactate method or consumes 1 umol NADH per minute in the pyruvate method under the stated conditions. |
| Molecular Weight | Approximately 38 kDa by SDS-PAGE |
| Source / Origin | Chicken heart |
| pH Range / Optimal pH | Stable at pH 4.0-10.0 for 24 hours at 25 degrees C; optimum pH 10.0-11.0 with lactate substrate and pH 7.0-8.0 with pyruvate substrate |
| Recommended Buffer System | Diluent: 50 mM potassium phosphate, 1 mg/mL BSA and 0.1% NaN3 at pH 7.0. Lactate method uses 350 mM CAPS pH 10.0, 1 M lithium lactate and 9 mM NAD. Pyruvate method uses 100 mM potassium phosphate pH 7.0, 25.4 mM pyruvate and 10 mM Tris-HCl containing 10 mg/mL NADH at pH 9.7. |
| Appearance | White amorphous powder |
| Alias | L-Lactate dehydrogenase |
| Product Type | L-lactate dehydrogenase enzyme raw material |
| Function / Efficacy | Catalyze reversible conversion between L-lactate and pyruvate with the NAD+/NADH cofactor pair. |
| Abbreviation | L-LDH |
| Customization | Specifications can be customized |
| Specific Activity | >=70 U/mg dry powder by the lactate method; >=100 U/mg dry powder by the pyruvate method |
