| Product Name | Bsu DNA Polymerase (Large Fragment, 50 U/μL) |
| Catalog No. | IVDRM-0273 |
| Description | Bsu DNA Polymerase (Large Fragment, 50 U/μL) is derived from Bacillus subtilis. The enzyme retains the 5'→3' polymerase activity of Bsu DNA polymerase I but lacks the 5'→3' exonuclease domain, and the large fragment itself is devoid of 3'→5' exonuclease activity. It is a DNA polymerase with strand-displacement activity for isothermal amplification, primarily applied in recombinase polymerase amplification (RPA) technology. |
| Intended Use | Core single-enzyme raw material for recombinase polymerase amplification (RPA) and other isothermal amplification applications. |
| Principle / Technology | Bsu DNA polymerase large fragment possesses inherent strand-displacement activity: during RPA, it extends invading primers while displacing the downstream DNA strand, enabling exponential DNA amplification at a constant low temperature (typically 37–42°C) without thermal cycling. |
| Detection Method | Recombinase polymerase amplification (RPA) and other isothermal amplification methods |
| Sample Type | DNA |
| Performance Range / Specifications | Concentration: 50 U/μL Retains 50% activity at 25°C—twice that of the Klenow fragment (3'→5' exo-) at the same temperature Lacks 3'→5' exonuclease activity and therefore cannot remove 3' unpaired overhangs; not suitable for generating blunt ends Storage buffer: 25 mM Tris-HCl, 50 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% glycerol, pH 7.4 @ 25°C Heat inactivation: 75°C for 20 min |
| Reaction Conditions / Protocol | Optimal for isothermal amplification at moderate temperatures; retains 50% activity at 25°C. Heat inactivation: 75°C for 20 min. |
| Components / Formulation | Supplied in storage buffer: 25 mM Tris-HCl, 50 mM NaCl, 1 mM DTT, 0.1 mM EDTA, 50% glycerol, pH 7.4 @ 25°C. |
| Storage Conditions | Store at -25 to -15°C. |
| Shelf Life | 2 years |
| Package Specifications | 2,500 U; 5,000 U; 50,000 U; 250,000 U |
| Product Form | Liquid |
| Quality Control | Purity and residual nuclease activity are tested for each lot. |
| Purity | High purity; verified by purity and residual nuclease assays. |
| Concentration | 50 U/μL |
| Source / Origin | Recombinantly expressed large fragment derived from Bacillus subtilis DNA polymerase I. |
| pH Range / Optimal pH | Storage buffer pH 7.4 @ 25°C |
| Expiration Date / Stability | Stable for 2 years when stored at -25 to -15°C. |
| Regulatory / Compliance | For research use and IVD raw material manufacturing. |
| Compatibility | Compatible with RPA reaction systems and other strand-displacement-based isothermal amplification workflows. |
| Application Notes / Precautions | Because it lacks 3'→5' exonuclease activity, the enzyme cannot excise 3' unpaired overhangs and is not suitable for blunt-end generation. |
| Heat Inactivation | 75°C for 20 min |
For research use only, not for clinical use.
