| Product Name | Antibody Hot-Start Taq DNA Polymerase (50 U/μL) |
| Catalog No. | IVDRM-0198 |
| Description | Antibody Hot-Start Taq DNA Polymerase (50 U/μL) is a mixture of a high-affinity anti-Taq monoclonal antibody and a thermostable recombinant wild-type Taq DNA polymerase. The antibody binds the polymerase so tightly that polymerase activity remains fully blocked even after incubation at 50°C for 30 minutes; heating at the pre-denaturation temperature for just 30 seconds irreversibly inactivates the antibody and releases full DNA polymerase activity. This hot-start mechanism effectively suppresses non-specific amplification caused by primer mis-annealing during reaction setup. The underlying Taq DNA polymerase is a thermostable recombinant enzyme with high template affinity, making it ideal for amplifying low-copy-number templates. Amplification products carry 3'-dA overhangs and can be cloned directly into T-vectors. Suitable for hot-start PCR and qPCR, including multiplex PCR. |
| Intended Use | For hot-start PCR and qPCR, including amplification of low-copy-number templates and multiplex PCR. |
| Principle / Technology | Antibody-mediated hot start: a high-affinity anti-Taq antibody blocks polymerase activity at low temperatures (blocking is maintained even at 50°C for 30 min). Heating at the pre-denaturation temperature for 30 sec fully inactivates the antibody and releases DNA polymerase activity, preventing non-specific amplification from primer mis-annealing during reaction setup. |
| Detection Method | PCR / qPCR amplification. |
| Performance Range / Specifications | Wide linear detection range: good linearity from 10^3 to 10^7 gene copies. No residual exonuclease activity. Suitable for multiplex PCR. High template affinity for efficient amplification of low-copy templates. Products carry 3'-dA overhangs for direct T-vector cloning. |
| Sensitivity / LOD | Good linearity across 10^3-10^7 gene copies; high template affinity supports amplification of low-copy-number templates. |
| Components / Formulation | Mixture of anti-Taq monoclonal antibody and thermostable recombinant wild-type Taq DNA polymerase, supplied at 50 U/μL. |
| Storage Conditions | Store at -25°C to -15°C. |
| Shelf Life | 2 years when stored at -25°C to -15°C. |
| Package Specifications | 100 μL; 1 mL; 25 mL |
| Product Form | Liquid enzyme formulation, 50 U/μL. |
| Concentration | 50 U/μL |
| Source / Origin | Thermostable recombinant wild-type Taq DNA polymerase complexed with a high-affinity anti-Taq antibody. |
| Application Notes / Precautions | The hot-start mechanism prevents non-specific amplification during reaction setup. Fidelity is comparable to conventional wild-type Taq DNA polymerase. Amplification products carry 3'-dA overhangs and can be cloned directly into T-vectors. |
| Heat Inactivation | The blocking antibody is fully inactivated by heating at the pre-denaturation temperature for 30 sec, releasing DNA polymerase activity. |
