| Product Name | Antibody Hot-Start B-Taq DNA Polymerase |
| Catalog No. | IVDRM-0098 |
| Description | Antibody Hot-Start B-Taq DNA Polymerase is a thermostable, mutant Taq DNA polymerase blocked by a specific antibody, providing markedly improved amplification sensitivity and specificity. Like wild-type Taq DNA polymerase, B-Taq possesses 5'-3' polymerase activity and 5'-3' exonuclease activity, but lacks 3'-5' exonuclease (proofreading) activity. B-Taq is a blood-tolerant DNA polymerase and exhibits excellent terminal A-addition characteristics, making it well suited for demanding PCR applications, including STR genotyping and probe-based assays. |
| Intended Use | Hot-start PCR amplification, including routine PCR, probe-based qPCR/PCR setups, and STR genotyping; for research use only. |
| Principle / Technology | Antibody-based hot-start mechanism: the polymerase is reversibly blocked by a specific antibody at room temperature, preventing non-specific amplification and primer-dimer formation during reaction setup. The antibody is denatured during the initial high-temperature pre-denaturation step, releasing fully active polymerase. B-Taq is a blood-tolerant, thermostable mutant Taq with 5'-3' polymerase and 5'-3' exonuclease activities and efficient A-addition. |
| Sample Type | DNA templates (1-20 ng per reaction recommended; approximately 1 ng for STR genotyping and approximately 20 ng for routine PCR); tolerant of blood-containing samples. |
| Performance Range / Specifications | Concentration: 5 U/μL Recommended usage: 2.5 U per 25 μL reaction Template range: 1-20 ng per 25 μL reaction |
| Specificity | Antibody-mediated hot start effectively suppresses non-specific amplification, providing high amplification specificity and sensitivity. |
| Reaction Conditions / Protocol | Recommended 25 μL reaction setup: 2× reaction buffer: 12.5 μL (1× final) Primer mix (10 μM): 0.2-1.25 μL (0.1-0.5 μM final) Probe mix (10 μM): 0.2-1.25 μL (0.1-0.5 μM final) Antibody Hot-Start B-Taq DNA Polymerase (5 U/μL): 0.5 μL (2.5 U per 25 μL) Template DNA: X μL (1-20 ng) Sterile ultrapure water: up to 25 μL Recommended two-step amplification program: Pre-denaturation: 95°C, 5 min, 1 cycle Denaturation: 95°C, 30 sec, 30 cycles Annealing/extension: 60°C, 1 min 30 sec Final extension: 60°C, 10-60 min, 1 cycle Note: DNA amount and primer concentrations above are recommended values and can be optimized for specific experiments. For STR genotyping, keep the template amount at approximately 1 ng; for routine PCR, use approximately 20 ng. Annealing/extension temperature and final extension time can be adjusted as required. |
