| Product Name | Anti-Taq DNA Polymerase Monoclonal Antibody (Polymerase Blocking) |
| Catalog No. | IVDRM-0282 |
| Description | This monoclonal antibody is designed for hot-start PCR. It binds to Taq DNA polymerase and inhibits its DNA polymerase activity at low temperatures, effectively preventing nonspecific primer annealing and primer-dimer-driven nonspecific amplification during reaction setup and low-temperature stages. The antibody is denatured during the initial DNA denaturation step of PCR, restoring full DNA polymerase activity and thereby achieving a hot-start effect. No special antibody inactivation treatment is required, and the product can be used under conventional PCR reaction conditions. |
| Intended Use | Raw material for hot-start PCR reagent formulation; used to block Taq DNA polymerase activity prior to thermal cycling for improved amplification specificity and sensitivity in molecular diagnostic and research PCR assays. |
| Principle / Technology | Antibody-mediated hot-start mechanism: the monoclonal antibody binds Taq DNA polymerase and blocks its 5' to 3' polymerase activity at low temperature. During the initial DNA denaturation step of PCR (95 °C), the antibody is heat-denatured and dissociates, releasing and restoring polymerase activity for specific amplification. |
| Detection Method | Hot-start PCR |
| Performance Range / Specifications | High detection sensitivity: the hot-start format facilitates detection of low-abundance target genes. Fast enzyme activity release: more than 95% of enzyme activity is released after heating at 95 °C for 20 s. Nuclease-free: free of exonuclease, endonuclease, and RNase contamination. |
| Sensitivity / LOD | The hot-start mechanism supports detection of low-abundance genes and provides higher detection sensitivity. |
| Specificity | Effectively suppresses nonspecific primer annealing and primer-dimer-induced nonspecific amplification at low temperatures. |
| Reaction Conditions / Protocol | At a concentration of 5 mg/mL, 1 uL of antibody is recommended to block 20-50 U of Taq DNA polymerase activity. Mix gently at room temperature for 20 min before use. Activity blocking efficiency is greater than 90%. |
| Components / Formulation | Monoclonal antibody solution against Taq DNA polymerase, supplied at 5 mg/mL. |
| Storage Conditions | Store at -25 °C to -15 °C. |
| Shelf Life | 2 years |
| Package Specifications | 100 µg 1 mg 5 mg 10 mg 100 mg 1000 mg |

| Concentration | 5 mg/mL |
| Activity / Unit Definition | 1 uL of antibody (5 mg/mL) blocks 20-50 U of Taq DNA polymerase activity; blocking efficiency > 90%. |
| Source / Origin | Monoclonal antibody against Taq DNA polymerase |
| Expiration Date / Stability | Stable for 2 years when stored at -25 °C to -15 °C. |
| Regulatory / Compliance | For use as an IVD raw material; for research and manufacturing use. |
| Compatibility | Compatible with conventional PCR reaction conditions; no special antibody inactivation step is required. |
| Application Notes / Precautions | Mix the antibody with Taq DNA polymerase at room temperature for 20 min before use to ensure complete blocking. |
| Recognition Site | Taq DNA polymerase (blocks 5' to 3' polymerase activity) |
| Heat Inactivation | The antibody is denatured during the initial DNA denaturation step of PCR; heating at 95 °C for 20 s releases more than 95% of enzyme activity. |
