| Product Name | Advanced Hot-Start Taq DNA Polymerase, Ultra-Clean (20 U/μL) |
| Catalog No. | IVDRM-0239 |
| Description | This product is a hot-start DNA polymerase dual-blocked with two proprietary monoclonal antibodies, which inhibit both the 5'->3' polymerase activity and the 5'->3' exonuclease activity of Taq DNA polymerase. Heating for 30 s at the pre-denaturation temperature fully inactivates the blocking antibodies, releasing both polymerase and exonuclease activities. The dual-blocking design not only effectively prevents non-specific amplification caused by mispriming or primer-dimers, but also suppresses fluorescence signal decay caused by probe degradation, making in vitro diagnostic reagents more stable during transportation or room-temperature handling. In addition, the enzyme is manufactured with an ultra-clean, low nucleic acid background process, resulting in extremely low residual nucleases and host genomic DNA. When combined with an optimized buffer system, it effectively reduces non-specific amplification caused by primer-probe interactions during mixing, reaction ramp-up, and long-term storage, supporting the development of fully pre-mixed real-time quantitative PCR (qPCR) systems. |
| Intended Use | Raw material enzyme for the development of in vitro diagnostic (IVD) qPCR reagents, including fully pre-mixed and multiplex real-time PCR systems. |
| Principle / Technology | Dual-antibody hot-start technology simultaneously blocks the 5'->3' polymerase and 5'->3' exonuclease activities; combined with an ultra-clean, low nucleic acid background manufacturing process that minimizes host-derived nucleic acid and nuclease residues. |
| Detection Method | Fluorescence-based real-time quantitative PCR (qPCR) |
| Performance Range / Specifications | E. coli genomic DNA residue <0.02 copies/100 U, among the lowest levels in the industry. Suitable for fully pre-mixed system development: 37 °C, 7-day full-premix stability validated in multi-primer/probe systems. Supports high-sensitivity assay development: the cleaner enzyme raw material reduces sequence complexity in the reaction system, improving detection sensitivity. |
| Sensitivity / LOD | Excellent amplification sensitivity; in a 4-plex qPCR system, the limit of detection reaches at least 250 copies/mL. |
| Specificity | Dual-antibody blocking effectively prevents non-specific amplification caused by mispriming or primer-dimers and suppresses fluorescence signal decay caused by probe degradation. No-template-control (NTC) testing in E. coli, Pseudomonas aeruginosa, and human genomic DNA background systems showed no detection in 48 NTC wells, confirming extremely low background DNA residue. |
| Reaction Conditions / Protocol | Hot-start activation: heat for 30 s at the pre-denaturation temperature to fully inactivate the blocking antibodies and release polymerase and exonuclease activities. |
| Components / Formulation | Hot-start Taq DNA polymerase supplied at 20 U/uL; recommended for use with a matched optimized buffer system (available separately). |
| Storage Conditions | Store at -25 to -15 °C. |
| Shelf Life | 2 years |
| Package Specifications | 1000 U; 10000 U; 25000 U; 100000 U |
| Product Form | Liquid enzyme solution, 20 U/uL |
| Quality Control | E. coli genomic DNA residue controlled at <0.02 copies/100 U; background DNA (E. coli, Pseudomonas aeruginosa, and human) verified as non-detected in 48-well NTC testing. |
| Purity | Ultra-low residual nucleases, host genomic DNA, and background bacterial DNA (ultra-clean, low nucleic acid background). |
| Concentration | 20 U/uL |
| Source / Origin | Recombinant Taq DNA polymerase expressed in E. coli |
| Expiration Date / Stability | Stable for 2 years at -25 to -15 °C; fully pre-mixed qPCR systems validated for 7-day stability at 37 °C. |
| Compatibility | Compatible with fully pre-mixed qPCR systems, multiplex (at least 4-plex) detection systems, and low-concentration sample detection. |
| Recommended Buffer System | Matched optimized buffer system (available separately) recommended to minimize non-specific amplification from primer-probe interactions. |
| Application Notes / Precautions | Supports qPCR systems with primers and probes pre-mixed in advance; ultra-low background DNA residue improves the reliability of low-copy and microbiome-related sample detection. |
| Batch-to-Batch Consistency | Strict residue control (<0.02 copies/100 U E. coli gDNA) applied across production batches. |
| Heat Inactivation | Not applicable (thermostable enzyme); the hot-start blocking antibodies are fully inactivated by 30 s heating at the pre-denaturation temperature. |
