Product Name 5 U/µL Exonuclease-Deficient Hot-Start Taq for Forensic Multiplex PCR
Catalog No. IEC-HMM-0305
Description Alta DiagnoTech antibody-modified hot-start Taq polymerase lacks 5'-to-3' exonuclease activity and remains strongly blocked below 70°C.
Intended Use For multiplex PCR, including forensic amplification workflows and primer/probe premix formulations.
Principle / Technology Site-directed mutation removes 5'-to-3' exonuclease activity, while a specific antibody blocks polymerase activity below 70°C. Heating at 95°C for 30 seconds releases more than 95% of enzyme activity.
Performance Range / Specifications Maintains strong activity blockade below 70°C and releases more than 95% activity after 30 seconds at 95°C.
Reaction Conditions / Protocol Activate at 95°C for 30 seconds. A final concentration of 0.2 µM per primer is recommended as a starting point.
Components / Formulation The enzyme is supplied at 5 U/µL with a 5× Taq buffer. The 1 kU, 10 kU and 100 kU formats contain 200 µL, 2 mL and 20 mL enzyme, respectively, with 1 mL, 10 mL and 100 mL buffer.
Storage Conditions Store at -25°C to -15°C.
Package Specifications Available in 1 kU, 10 kU and 100 kU formats.
Concentration 5 U/µL
Activity / Unit Definition One unit incorporates 10 nmol of deoxynucleotide into DNA in 30 minutes at 72°C using M13 mp18 single-stranded virion DNA as template-primer.
Shipping Conditions Ship at 0°C or below.
Compatibility Designed for multiplex PCR and forensic testing.
Application Notes / Precautions Validate each primer by PCR and calibrate the actual concentration of newly synthesized primers. Increase the corresponding primer amount moderately if a target produces low yield.
Product Type Exonuclease-deficient antibody-modified hot-start Taq DNA polymerase
Function / Efficacy Reduces nonspecific amplification and improves primer/probe premix stability, specificity and low-template sensitivity.

For research use only, not for clinical use.
