Description Alta DiagnoTech beta-galactosidase is a white amorphous enzyme powder characterized for ONPG-based biochemical reagent development.

Principle / Technology Beta-galactosidase hydrolyzes o-nitrophenyl beta-D-galactoside, and product formation is monitored at 405 nm.
Detection Method Kinetic ONPG absorbance assay at 405 nm
Performance Range / Specifications Specific activity is at least 40 U/mg; optimal assay range is 100-1,000 U/L.
Reaction Conditions / Protocol Preincubate 0.520 mL assay reagent with 0.020 mL diluted enzyme for 1 minute at 37 degrees C, add 0.060 mL of 22.7 mM ONPG, react for 4 minutes at 37 degrees C and calculate the A405 change from minute 3 to minute 4. Total reaction volume is 0.6 mL.
