Product Name >=150 U/mg Lactate Dehydrogenase for 340 nm ALT/AST Assays
Catalog No. IEC-HMM-0222
Description Alta DiagnoTech lactate dehydrogenase is a white amorphous enzyme powder characterized for NADH-based activity testing and use in alanine and aspartate aminotransferase reagent development.
Intended Use Enzyme raw material for alanine aminotransferase and aspartate aminotransferase biochemical assay reagents.
Detection Method Kinetic absorbance measurement at 340 nm
Performance Range / Specifications Specific activity >=150 U/mg; assay linear range 100-1,000 U/L; recommended sample range 500-1,000 U/L.
Reaction Conditions / Protocol Prepare Reagent 1 with 12.1 g Tris in 800 mL deionized water, adjust to pH 7.70 at 25 degrees C with HCl, dissolve 1 g sodium azide and bring to 1 L; immediately before use, add sodium pyruvate to 0.4 g/L. Prepare Reagent 2 with 21.1 g CAPS in 800 mL water, adjust to pH 9.70 with NaOH, dissolve 1.20 g NADH and 1 g sodium azide and bring to 1 L. For the assay, combine 1.00 mL Reagent 1 and 0.020 mL enzyme solution, preincubate for 60 seconds at 37 degrees C, add 0.200 mL Reagent 2, react for 1 minute, then determine the 1-minute absorbance-change rate at 340 nm. Activity in U/L equals Delta A/min x 9,807 x dilution factor.
Storage Conditions Store at -20 degrees C; prepared assay reagents are stored at 2-8 degrees C.
Shelf Life At least 1 year at -20 degrees C; prepared Reagent 1 and Reagent 2 are usable for 1 week at 2-8 degrees C.
Product Form White amorphous powder
Activity / Unit Definition One unit is the amount of enzyme that generates 1 umol NAD+ per minute under the stated reaction conditions.
Molecular Weight 42 kDa
pH Range / Optimal pH Stable at pH 4.5-10.0 for 24 hours at 25 degrees C; optimum pH 6.5-8.0
Recommended Buffer System Sample diluent: dissolve 1 g BSA and 1.36 g potassium dihydrogen phosphate in 800 mL deionized water, adjust to pH 7.0 at 25 degrees C with NaOH and bring to 1 L. The diluent is stable for 2 months at 2-8 degrees C.
