Description Alta DiagnoTech cholesterol esterase is an off-white amorphous enzyme powder for total cholesterol, HDL-C and LDL-C biochemical reagent development.
Intended Use Enzyme raw material for total cholesterol, HDL cholesterol and LDL cholesterol biochemical assay kits.
Detection Method Cholesterol-oxidase/peroxidase-coupled absorbance assay at 505 nm
Performance Range / Specifications Specific activity >=15 U/mg; assay linear range 100-300 U/L.
Reaction Conditions / Protocol Use potassium phosphate reagent at pH 7.0 containing 4-aminoantipyrine, peroxidase and cholesterol oxidase, together with phenol and a cholesterol-linoleate/Triton X-100/sodium-cholate substrate. Combine 0.3 mL reagent 1, 0.45 mL phenol reagent and 0.45 mL substrate, preincubate for 180 seconds at 37 degrees C, add 0.04 mL sample and determine the one-minute rate after 2 minutes at 505 nm.
