Reaction Conditions / Protocol Use potassium phosphate at pH 7.5 with ATP, MgCl2, CoA and palmitic acid in Triton X-100. Incubate 320 uL cofactor reagent plus 80 uL substrate for 1 minute at 37 degrees C, add 20 uL sample and react for 10 minutes. Add 800 uL color reagent and 20 uL phenol, incubate for 2 minutes, add 20 uL acetyl-CoA oxidase, react for 5 minutes at 37 degrees C and read at 500 nm against a dilution-buffer blank.
