| Catalog No. | IEC-HMM-0260 |
| Description | Alta DiagnoTech uricase is a pale-yellowish white amorphous enzyme powder for uric-acid biochemical reagent development. |
| Intended Use | Enzyme raw material for uric acid biochemical assay reagents. |
| Detection Method | Peroxidase-coupled kinetic colorimetric assay at 505 nm |
| Performance Range / Specifications | Specific activity >=1.0 U/mg; assay linear and recommended sample range 400-600 U/L. |
| Reaction Conditions / Protocol | Prepare Reagent 1 by dissolving 6.06 g Tris in 800 mL water, adjusting to pH 8.50 with HCl, adding 6.0 g phenol, 0.3462 g EDTA disodium salt and 1 g sodium azide, adjusting to pH 8.50 +/- 0.05 at 25 degrees C and bringing to 1 L; store for up to 2 weeks at 2-8 degrees C. Prepare Reagent 2 in 700 mL water with 0.8 g lithium carbonate, 30.1 g disodium hydrogen phosphate dodecahydrate, 0.884 g pre-dissolved oxalic acid and 2.5 g sodium dihydrogen phosphate dihydrate; adjust to pH 7.50, add 0.25 g uric acid, 2.34 g 4-aminoantipyrine, 6.0 kU peroxidase and 1 g sodium azide, then bring to 1 L at pH 7.50 +/- 0.05. Combine 0.500 mL Reagent 1 and 2.500 mL Reagent 2, equilibrate at 37 degrees C, add 0.030 mL enzyme sample after 4 minutes, then after 2 minutes determine the 1-minute absorbance-change rate at 505 nm. Activity in U/L equals Delta A/min x 15,188 x dilution factor. |
