| Reaction Conditions / Protocol | Use Tris buffer at pH 8.0 with 4-aminoantipyrine, phenol, peroxidase, Triton X-100 and palmitoyl-CoA. Combine 0.2 mL buffer, 0.1 mL each of 4-AA, phenol, peroxidase, Triton X-100 and substrate plus 0.3 mL water; preincubate for 300 seconds at 37 degrees C, add 0.02 mL sample and determine the one-minute rate at 500 nm. The enzyme dilution buffer contains FAD and ATP. |
