Product Name Universal TaqMan Pro U+ qPCR Mix (One Tube)
Catalog No. RESK-0115
Description A multiplex fluorescent quantitative PCR (qPCR) master mix built on a next-generation antibody-based hot-start Taq DNA polymerase. This universal, fully pre-mixed one-tube reagent delivers excellent amplification performance with the majority of primer-probe sets, supports primers and probes pre-mixed in advance, and is compatible with fast-cycling programs (detection time 35 min or less). The 2x mix contains factors that suppress non-specific PCR amplification and enhance multiplex amplification efficiency, improving both sensitivity and specificity. A dUTP/UDG carryover contamination control system, in which dUTP replaces dTTP, effectively prevents aerosol contamination and ensures the accuracy of detection results.
Intended Use For DNA-template multiplex real-time quantitative PCR (qPCR) amplification in research and molecular diagnostic assay development; validated across pathogen, human, and plant targets.
Principle / Technology TaqMan probe-based real-time quantitative PCR using a next-generation antibody-based hot-start Taq DNA polymerase. The dUTP/UDG anti-contamination system substitutes dUTP for dTTP so that uracil-DNA glycosylase (UDG) degrades carryover amplicons before amplification, eliminating aerosol contamination risk.
Detection Method Real-time fluorescence detection with TaqMan hydrolysis probes; supports multiplex (3-plex and 4-plex validated) amplification.
Sample Type DNA templates; validated on pathogen, human-derived, and plant targets.
Performance Range / Specifications Universal compatibility: excellent amplification performance with the majority of primer-probe sets.
Full premix stability: amplification performance remains unchanged after 14 days of accelerated storage at 37 °C with primers and probes pre-mixed.
Fast-cycling support: detection time 35 min or less (results in 30 min under a fast program).
2x mix formulation contains factors that suppress non-specific PCR amplification and enhance multiplex qPCR efficiency, increasing multiplex sensitivity and specificity.
dUTP/UDG anti-contamination system prevents aerosol contamination and ensures result accuracy.
Sensitivity / LOD Amplification efficiency above 99% across 7 gradient samples from 1 to 10^6 copies/uL under both standard and fast programs.
Specificity The 2x mix contains factors that effectively suppress non-specific PCR amplification, improving the specificity of multiplex amplification.
Reaction Conditions / Protocol Supports both standard (approx. 1 h 6 min) and fast (30 min) cycling programs. Fast program maintains amplification efficiency above 99%, with Ct deviation (delta Ct) within +/-0.5 and fluorescence deviation within +/-10% versus the standard program; amplification curves show a typical sigmoidal (S) shape. Validated on standard real-time PCR instruments.
Components / Formulation 2x one-tube premix containing antibody-based hot-start Taq DNA polymerase, dNTPs (dUTP in place of dTTP), UDG, reaction buffer, and additives that suppress non-specific amplification and enhance multiplex amplification efficiency.
Storage Conditions Store at -25 to -15 °C.
Shelf Life 18 months.
Package Specifications 100 T; 1,000 T; 10,000 T; 15,000 T; 20,000 T
Product Form 2x liquid premix (one-tube master mix).
Expiration Date / Stability Valid for 18 months when stored at -25 to -15 °C. Full premix shows no change in amplification performance after 7 days at 4 °C or 37 °C, after 50 freeze-thaw cycles, or after 14 days of accelerated storage at 37 °C; Ct deviation (delta Ct) versus the -20 °C control remains within +/-0.5 and fluorescence deviation within 15% across 3-plex and 4-plex amplification systems.
Compatibility Compatible with the majority of primer-probe sets and with standard and fast-cycling programs; validated on standard real-time PCR instruments.
Application Notes / Precautions Primers and probes can be pre-mixed with the reagent in advance without loss of performance. Use UDG pretreatment to eliminate carryover contamination from previous dUTP-containing amplicons.
